摘要
目的探讨Synphilin-1蛋白对小鼠黑质纹状体多巴胺能神经元递质传递的影响。方法利用转基因技术制备Synphilin-1转基因小鼠(转基因组,11只),另以野生型小鼠为非转基因组(11只)作对照。通过加速转圈测试和足迹分析观察两组小鼠的运动能力。分别测定两组小鼠的多巴胺及其代谢产物,并且测定参与纹状体多巴胺代谢酶的表达水平,进行分析比较。结果转基因组小鼠的运动能力较非转基因组下降(转圈能力减弱、步长较短),且转基因组小鼠表现为短暂步态。Synphilin-1蛋白使黑质纹状体多巴胺能神经元的多巴胺代谢产物/多巴胺的比例明显下降(P<0.05),而参与多巴胺代谢的酶的含量并无明显变化(P>0.05)。结论 Synphilin-1蛋白引起小鼠运动能力损害,对黑质纹状体多巴胺代谢影响主要是通过改变突触前膜多巴胺能神经元递质传递来实现的。
Objective To investigate the effect of synphilin-1 on the metabolism of dopamine in dopaminergic neurons. Methods Rotarod performance test and step length test were conducted in 11 synphilin-l-transgenic mice and 11 non-transgenic mice (controls). The levels of dopamine, dopamine metabolites and related enzymes were measured. Results The motor function was exhibited in synphilin-l-transgenic mice (P 〈0.05). The enzyme activities related to dopamine metabolism in transgenic mice were not changed compared to control group (P ~0.05). Conclusion Synphilin-1 is involved in motor functional impairments, which may be associated with the release and repackage of dopamine at the presynaptic membrane of dopaminergic neuron.
