摘要
目的观察尾静脉放血对足底电击大鼠焦虑样行为及海马区炎症因子的影响。方法 SD大鼠(18只)按体重分层随机分为正常组、模型组及放血组,予以足底电击(0.8m A,2-25s随机发生1次电击,共5min,1次/天,7天)结合孤养复制焦虑模型,放血组在造模前尾静脉放血(15滴,1次),7天后以高架十字迷宫实验评价焦虑行为,Elisa检测海马区炎症因子。结果放血对焦虑行为的影响:模型组进入闭臂的次数及时间百分比显著高于正常组(P<0.05),放血组进入闭臂的次数百分比显著低于模型组(P<0.05)。放血对海马炎症因子的影响:模型组海马区炎症因子(TNF-α,IL-1β,IL-6)明显高于正常组(P<0.05),放血组炎症因子明显低于模型组(P<0.05)。结论放血对焦虑行为的影响:模型组进入闭臂的次数及时间百分比显著高于正常组(P<0.05),放血组进入闭臂的次数百分比显著低于模型组(P<0.05)。放血对海马炎症因子的影响:模型组海马区炎症因子(TNF-α,IL-1β,IL-6)明显高于正常组(P<0.05),放血组炎症因子明显低于模型组(P<0.05)。
Objective This study was to investigate the effects of bloodletting on reducing stress-induced anxiety-like behavior and expression of hippocampus inflammatory cytokines. Methods Rats( 18) were randomized to 3 groups of control,model and bloodletting group. In the model and bloodletting groups,stress was produced by inevitability footshock( 0. 8m A,random occurred once in 2 to 25 seconds,lasted for 5 minutes,once a day,7days in total) combined with separation. An hour before footshock,the bloodletting group got bloodletting with surgical blade prickedrat caudal vein( 15 drops,once). Anxiety-like behavior was evaluated by elevated plus maze on the 8th day. Elisawas used to detect hippocampus inflammatory cytokines. The control group did not get any processing. Results Model group showed significantly enhanced anxiety-like behavior as compared to control( P〈0. 05),bloodletting could attenuate it( P〈0. 05). The hippocampus inflammatory cytokines( TNF-α,IL-1β,IL-6) were increased significantly after exposure to stress( P〈0. 05). Pre-treatment with the caudal vein bloodletting attenuated the increased inflammatory cytokines( IL-1β,IL-6) levels instressed rats( P〈0. 05). Conclusion Anxiety-like behavior accompanied with the increased inflammatory cytokines,bloodletting reduced anxiety-like behavior,which may attribute to the decrease of hippocampus inflammatory cytokines after bloodletting.
