摘要
目的探讨红景天苷(Salidroside,Sal)对大鼠颅脑外伤的神经保护作用及其机制。方法健康成年SD大鼠36只,随机分为3组:假手术组(Sham组)、对照组(TBI组)、Sal给药组(TBI+Sal组)。通过Feeney's自由落体打击法制作大鼠颅脑损伤模型,24 h后对大鼠神经功能损伤评分,对神经元行尼氏(Nissl)染色,检测动物损伤脑组织中GSH、SOD及MDA的水平,Tunel染色检测细胞凋亡。结果 TBI+Sal组较TBI组,降低了神经功能缺损评分、细胞凋亡数量、MDA水平,显著增加了Nissl阳性细胞的数量、SOD的活性以及GSH的水平。结论 Sal能减轻大鼠TBI后神经功能缺损,具有神经保护作用,此作用依赖于抗氧化及抗凋亡的作用机制。
Objective The potential protection of salidroside on adult rats against the traumatic brain injury( TBI) was investigated.Methods Thirty-six adult rats were randomly divided into three groups,ie,sham group,TBI group and TBI + Sal group.We employed Feeney 's weight-drop model to ascertain whether intraperitoneal administration of salidroside( 40 mg / kg) post TBI could reduce the severity of neurological deficits and hippocampal neuron loss.The levels of malondialdehyde( MDA) and glutathione( GSH),the activity of superoxide dismutase( SOD),and the number of terminal deoxynucleotidyl transferase-mediated d UTP nick end labeling( TUNEL)-positive apoptotic cells were also measured to characterize the antioxidative and antiapoptotic properties.Results A significant reduction of neurological deficits and hippocampal neuron loss was observed in the TBI + sal group( 40 mg/kg) at 24 h after TBI compared with the TBI group.Furthermore,administration of salidroside( 40 mg/kg)significantly increased the activity of SOD,the level of GSH,and also reduced the level of MDA and the number of apoptotic cells at 24 h after TBI.Conclusion In summary,these results suggests that salidroside plays a neuroprotective effect against TBI,which may be associated with its antioxidative and antiapoptotic functions.
