摘要
目的:考察家蚕丝素对凝血酶的固定化作用。方法:以家蚕丝素为载体,戊二醛为交联剂制备固定化凝血酶。以固定化凝血酶的活力回收率(AR)为考察指标,采用正交试验确定丝素固定化凝血酶的最佳工艺条件。结果:丝素固定化凝血酶的最佳工艺条件为:固定时间为6 h,加酶量为2 400 NIH·g^(-1)载体,温度为25℃,p H为7.6。固定化凝血酶的回收率(AR)达67.22%。结论:丝素对凝血酶有良好的固定化作用。
Objective: To investigate the effects of silk fibroin on the immobilization of thrombin. Methods: The immobilized thrombin was prepared using silk fibroin as the carrier and glutaraldehyde as the crosslinking agent. With activity yield as the index,the process conditions of silk fibroin immobilized thrombin were determined by an orthogonal test. Results: The optimum process conditions of immobilized thrombin treated with silk fibroin were as follows: the immobilization time was 6 h,the enzyme dosage was 2 400 NIH·g^(-1)casein,the temperature was 25℃ and p H was 7.6. The activity recovery of immobilized thrombin was 67.22%. Conclusion: Silk fibroin has the positive immobilization effect on thrombin.
