黄芪多糖抑制NF-κB/MAPK信号通路和改善哮喘大鼠气道炎症的作用 被引量:88

Astragalus polysaccharide regulates NF-κB/MAPK signaling pathway and attenuates airway inflammation in OVA-induced asthmatic rats

摘要 目的观察黄芪多糖(astragalus polysaccharide,APS)对哮喘大鼠气道炎症及NF-κB/MAPK信号通路的影响。方法采用卵蛋白(ovalbumin,OVA)致敏法制备哮喘模型并予以药物治疗。观察支气管灌洗液(bronchoalveolar lavage fluid,BALF)中炎症细胞分类计数、肺组织病理变化、肺泡Ⅰ型上皮超微结构变化;测定肺组织NF-κB p65、磷酸化NF-κB p65(p-NF-κB p65)、磷酸化IκBα(p-IκBα)、ERK1/2、磷酸化ERK1/2(p-ERK1/2)、JNK、磷酸化JNK(p-JNK)、p38 MAPK、磷酸化p38 MAPK(p-p38 MAPK)、IL-1β、IL-4、IL-5、IL-6及IL-13含量的变化。结果与正常组比较,OVA致敏明显升高哮喘组炎症细胞数量、增强NF-κB/MAPK信号通路活性。APS可改善哮喘大鼠气道炎症、肺Ⅰ型上皮损伤,并明显抑制NF-κB/MAPK信号通路活性。结论 APS改善哮喘大鼠气道炎症、细胞损伤的机制可能与抑制NF-κB/MAPK信号通路有关。 Aim To detect the possible ameliorative effects of APS on the airway inflammation and whether the effects are associated with inhibiting the NF-κB /MAPK signaling pathway in ovalbumin( OVA)-induced asthmatic rats. Methods Asthma was induced by OVA sensitization and challenge. The asthmatic rats in the APS group were treated with APS. Pulmonary inflammation was assessed with hematoxylin and eosin staining and inflammatory cell counting in BALF. Ultrastructural changes of type I pneumocyte were observed by electron microscopy. Levels of NF-κB p65,p-NF-κB p65,p-IκBα,ERK1 /2,p-ERK1 /2,JNK,p-JNK,p38 MAPK,p-p38 MAPK,IL-1β,IL-4,IL-5,IL-6 and IL-13 were measured using ELISA to assess the activity of NF-κB / MAPK signaling pathway.Results Compared to the normal group,OVA induced significantly pulmonary inflammation and ultrastructural changes of type I pneumocyte in the asthma group. Besides,OVA increased the activity of the NF-κB / MAPK signaling pathway and promoted the production of inflammatory cytokines IL-1β,IL-4,IL-5,IL-6and IL-13 in the asthma group. If compared to the asthma group,APS markedly attenuated the pulmonary inflammation and type I pneumocyte damage,as well as inhibited the activity of the NF-κB / MAPK signaling pathway and decreased the production of inflammatory cytokines in the APS group. Conclusion The ameliorative effects of APS on airway inflammation might be associated with the inhibition of the activity of the NF-κB /MAPK signaling pathway.
