摘要
目的应用小干扰RNA(siRNA)干扰肾癌786-0细胞及ACHN细胞Cullinl(Cull)基因的表达,探讨Cull基因在人肾癌细胞增殖、迁移和侵袭中的作用。方法用CullsiRNA和ControlsiRNA转染人肾癌786-0及ACHN细胞,转染24h后行CCK-8细胞增殖实验,转染48h后行迁移与侵袭实验,转染48h后提取蛋白,Westernblot检测Cull蛋白及基质金属蛋白酶2(MMP-2)和MMP-9蛋白的表达。结果转染CullsiRNA后,肾癌786-0及ACHN细胞的Cull表达下降明显,肾癌细胞的增殖能力、迁移能力及侵袭能力下降,MMP-2及MMP-9表达下降。结论干涉Cull基因可以抑制肾癌786-0细胞及ACHN细胞的增殖,并通过降低肾癌细胞内MMP-2和MMP-9的表达来抑制肾癌细胞迁移和侵袭。
Objective To investigate the effects of Cullinl ( Cull ) gene on the proliferation, migration and invasion of human renal carcinoma cells. Methods Both CullsiRNA and control siRNA sequence was synthesized chemically in vitro and transfected into 786 - O and ACHN renal cells. After 24 hours of transfection, the proliferation of the ceils was determined by CCK8 assay. After 48 hours of transfection, migration and invasion assays were performed. Meanwhile, the levels of Cull, and MMP - 2 and MMP - 9 were detected by Western blotting. Results After RNA interference, the level of Cull in 786 -O and ACHN cells was remarkably decreased. Knockdown of Cull suppressed the proliferation, in- vasion and migration of the ceils. The amounts of MMP - 2 and MMP - 9 were also decreased. Conclusions Decreased expression of Cull can inhibit the proliferation of 786 - O and ACHN cells and suppress their migration and invasion through down - regulation of MMP - 2 and MMP - 9.
