摘要
目的了解2010-2012年广州地区腹泻患者的博卡病毒的分子变异情况。方法采用普通RT-PCR方法扩增2010-2012年14例人博卡病毒全基因组序列并测序,利用生物信息学软件分别对测序标本的全基因组和编码基因NS1、NP1和VP1/VP2序列进行比对和分析,了解广州地区人博卡病毒的进化变异特征。结果 14例HBo Vs全基因组长约5.2 kb,包含3个开放阅读框(ORF),编码3种蛋白:NS1、NP1、VP1/VP2。其中NS1基因高度保守,VP1/VP2有一些变异,但基于VP1/VP2序列进行的进化分析得到的结果与全基因组分析得到的结果一致。所有14例HBo V病毒株同属于HBo V1型,且没有发生明显的变异。结论2010-2012年广州地区分离到的14例人博卡病毒株同属于HBo V1型,没有发生明显的变异;NS1基因高度保守,VP1/VP2变异率相对较高,且进化分析结果与全基因组进化分析结果一致,可用于监测HBo V的变异。
Objectives To better understand the molecular evolution characteristic of human bocavirus in diarrhea patients in Guangzhou from 2010 to 2012. Methods The complete genomes of 14 human bocavirus were amplified by RT-PCR and sequenced. The sequences were then analyzed by relative bioinformatics software to find out the molecular evolution characteristic of human bocavirus in diarrhea patients in Guangzhou from 2010 to 2012. Results The length of human bocavirus complete genomes is about 5.2 kb, consists of three open reading frames(ORF) : NS1, NP1, VP1 or VP2.The NS1 gene is highly conserved, while VP1 / VP2 has relatively larger variation. Evolution analysis result based on VP1/ VP2 sequences are consistent with genome-wide sequence analysis. All 14 cases of human bocavirus strains belong to HBo V1 and no obvious variation was found. Conclusion All 14 cases of human bocavirus strains isolated in Guangzhou from 2010 to 2012 belong to HBo V1 and no obvious variation was found. NS1 gene is highly conserved, while VP1 /VP2 mutation rate is relatively high and can be used for monitoring the variation of human bocavirus.
