摘要
目的:通过分别观察脾虚痰浊证大鼠胃、心肌及下丘脑组织c AMP/PKA-Gp信号转导通路相关蛋白的变化,探讨脾虚痰浊证的改变与c AMP/PKA-Gp信号转导通路的相关性。方法:16只SPF级雄性SD大鼠,随机分为正常组和脾虚痰浊组,每组各8只,脾虚痰浊证模型采用喂养高脂饲料,时间6个月。造模结束后,首先通过大鼠的一般状态和生理情况的改变进行模型评价,再分别取胃、心和下丘脑组织。分别采用ELISA法检测痰浊证大鼠不同组织c AMP活性,采用实时荧光定量PCR法和Western blot法检测PKA、CREB、PHK和Gp mRNA和蛋白的表达情况。结果:与正常组大鼠比较,脾虚痰浊证大鼠胃、心及下丘脑组织c AMP活性和PKA、CREB mRNA和蛋白的表达均有所减弱(P<0.01或P<0.05);并且脾虚痰浊证大鼠胃和心组织Gp mRNA和蛋白的表达也均有所减弱(P<0.01或P<0.05);此外,脾虚痰浊证大鼠心组织的PHK mRNA和蛋白的表达与正常组比较也存在显著性差异(P<0.01)。结论:脾虚痰浊证大鼠模型成立后,其胃、心及下丘脑组织的改变均与c AMP/PKA-Gp信号转导通路存在一定的相关性。
Objective: To discuss the changes of c AMP / PKA- Gp signal transduction pathway in the gastric and cardiac muscle and hypothalamus tissue in spleen deficiency with phlegm turbidity rats and probe the relevance of the changes of spleen deficiency with phlegm turbidity syndrome and c AMP / PKA- Gp signal transduction pathway. Methods: A total of 16 male SD SPF rats were randomly assigned into two groups,including control group and spleen deficiency with phlegm turbidity group. The modeling method of spleen deficiency with phlegm turbidity rats was reared with high fat diet for 6 months. The evaluation method of modeling was used for observing the general state and physiological conditions of the rats. We obtained the gastric and cardiac muscles and hypothalamus tissue. And then we tested the c AMP activity and PKA,CREB,PHK and Gp mRNA and protein expression of the gastric tissue. Results: The gastric and cardiac muscles and hypothalamus tissue c AMP activity and PKA,CREB mRNA and protein expression of spleen deficiency with phlegm turbidity rats were weakened compared with those of the control group( P 〈0. 01 or P 〈 0. 05). The gastric and cardiac muscle tissue’ s Gp mRNA and protein expression of spleen deficiency with phlegm turbidity rats were weakened( P 〈 0. 01 or P 〈 0. 05). In addition,the cardiac muscle tissue’s PHK mRNA and protein expression of spleen deficiency with phlegm turbidity rats were weakened( P 〈 0. 01). Conclusion: After building the modeling of spleen deficiency with phlegm turbidity rats,there is relevance between the changes of c AMP / PKA- Gp signal transduction pathway in the gastric and cardiac muscles and hypothalamus tissue.
