摘要
目的:观察脾虚痰浊证大鼠主动脉c AMP/PKA信号通路变化,探讨其信号通路变化与脾虚痰浊证的关系。方法:16只SD大鼠,每组8只,随机分为空白对照组、脾虚痰浊组。全自动生化分析仪检测血清TC、TG、LDL-C、HDLC水平,ELISA法检测血浆中c AMP浓度,RT-PCR和Western blot检测肝脏GP、CREB、PKA和PHK蛋白表达水平。结果:与空白对照组相比,TG含量显著增高,主动脉c AMP活性显著降低,GP、CREB、PKA、PHK mRNA含量显著下降,GP、CREB、PKA、PHK蛋白表达显著下降。结论:脾虚痰浊证大鼠主动脉c AMP/PKA信号通路的改变可能与调控血脂代谢相关基因GP、CREB、PKA、PHK的表达有关。
Objective : To observe the aorta cAMP/PKA signal pathway changes in the rats with spleen phlegm and discuss the relationship between the signal pathway changes and spleen phlegm. Methods:Sixteen SD rats were randomly divided into con- trolled group and spleen phlegm group with eight in each. Detect serum TC, TG, LDL - C and HDL - C levels with an automatic biochemical analyzer. Detect cAMP concentrations in the plasma with the ELISA method. Apply the RT - PCR and Western blot to detect CREB, GP, PKA and PHK protein content levels in the liver. Results : Compared with the controlled group, the TG levels were significantly increased and aortic cAMP levels were significantly decreased. GP,CREB,PKA and pI-IK mRNA were signifi- cantly decreased and GP, CREB, PKA and PHK protein contents were significantly decreased. Conclusion:Aorta cAMP/PKA sig- nal pathway changes in the rats with spleen phlegm may be associated with the regulation of lipid metabolism genes GP, CREB, PKA and PHK.
