摘要
目的:探讨锌指E-盒结合同源异形盒1(ZEB1)在宫颈癌组织和细胞中的表达及其与宫颈癌发生、发展的关系。方法:应用免疫组织化学、免疫细胞化学、蛋白免疫印迹和RT-PCR法检测65例宫颈鳞癌、31例宫颈上皮内瘤变、40例正常宫颈组织以及4株宫颈癌细胞系He La、Si Ha、C33-A、Caski中ZEB1的表达情况,并分析其与宫颈癌临床及病理特征的关系。结果:免疫组化结果显示ZEB1在宫颈鳞癌、宫颈上皮内瘤变及正常宫颈组织中的阳性表达率依次降低,分别为81.5%、77.4%、30%(P<0.05)。同时,RT-PCR和蛋白免疫印迹分析显示ZEB1在宫颈癌组织中的表达较正常宫颈组织高,具有统计学差异(P<0.05)。ZEB1的表达情况与宫颈癌的病理分化程度、临床分期和是否淋巴结转移有关(P<0.05),与患者年龄无关(P>0.05)。另外,免疫细胞化学、蛋白免疫印迹和RT-PCR显示ZEB1在宫颈癌细胞系中均有不同程度的表达。结论:ZEB1的异常表达与宫颈癌的发生、发展密切相关,其可能促进宫颈癌的浸润和转移进程。
Objective :To investigate the expression of zinc finger E -box binding homeobox 1 (ZEB1) in human cervical carcinoma tissue and its role in the occurrence and development of cervical cancer. Methods:Immunohistochemistry,immunocytochemistry,Western Blot and RT - PCR were used to detect the expression of ZEB1 in 65 cases of cervical carcinoma,31 cases of cancer in situ,40 cases of normal cervix tissue and 4 cervical cancer cell lines, such as HeLa,SiHa, C33 -A and Caski. The relationship between ZEB1 and clinical and pathological features of cervical carcinoma were analyzed. Results:Immunohistochemistry results showed ZEB1 had a decrease positive rates in cervical carcinoma,cancer in situ and normal cervix,which were 85. 1% ,77.4% and 30% ,respectively (P 〈 0.05). There was significant difference in expression of ZEB1 in various degree of histological differentiation, clinical stage and lymph node metastasis situation (P 〈 0.05 ) ,which was opposite to age (P 〉 0.05 ). Moreover, RT - PCR results showed the expression level of ZEB1 in cervical carcinoma was higher than normal cervical tissue (P 〈 0.05). Immunocytochemistry, Western Blot and RT - PCR results showed different expression level of ZEB1 in different cervical cancer cell lines and the expression in cervical cancer was higher than normal cervical tissue ( P 〈 0.05 ). Conclu- sion : The abnormally high expression of ZEB1 is closely related to the occurrence and development of cervical cancer, which may play an important role in the process of cancer invasion and metastasis.
