PEG-rhG-CSF预处理对BUCY方案致小鼠骨髓抑制模型骨髓造血恢复的影响 被引量:7

摘要 目的:研究聚乙二醇化粒细胞集落刺激因子(PEG-rh G-CSF)预处理后不同时间间隔对BUCY方案致小鼠骨髓抑制模型骨髓及外周血象恢复情况的影响,为进一步扩展PEG-rh G-CSF的临床应用范围提供理论依据。方法:BLAB/c雌性小鼠50只,随机分为5组(n=10)。实验组(Group6、Group10、Group13、Group16)皮下注射PEG-rh G-CSF 70mg·kg^(-1),给药后的第6、10、13及16天给予白消安(BU)35mg·kg^(-1)·d^(-1)连续4天,之后给予环磷酰胺(CY)50mg·kg^(-1)·d^(-1)连续2天。粒细胞缺乏组(Model)给予BU 35mg·kg^(-1)·d^(-1)连续4天,之后给予CY 50mg·kg^(-1)·d^(-1)连续2天。结果:经BUCY方案处理后,除Group6组白细胞于第5天开始下降外,其余各组白细胞均于第3天开始下降;各组的(Model、Group6、Group10、Group13、Group16)白细胞最低值分别出现在第11、9、9、11及11天,白细胞恢复正常的时间分别为第19、23、23、21及19天。第19天时Model和Group16组的骨髓病理结果均已趋于正常,其他组骨髓组织还存在不同程度的空泡结构,表现为低增生骨髓状态。结论:使用PEG-rh G-CSF后16天对于BUCY方案引起的小鼠骨髓抑制模型的骨髓恢复已无影响,临床5天用药的3周化疗方案可以尝试性使用PEG-rh G-CSF预防粒细胞降低。 Objective: To research ted by busulfan and cyclophosphamid BLAB/c female mice were randomly the effect of bone marrow and peripheral blood of myelosuppresived mice treae(BUCY) applied by PEG- rhG- CSF in different time intervals. Methods: divided into 5 groups ( n = 10 ) , the mice in the test groups ( Group6, Group10 , Group13 ,Group16 )were injected subcutaneously by PEG- rhG- CSF 70mg ·kg^-1, and then treated by BU 35mg ·kg^-1· d^-1 for 4 days and CY 50mg ·kg^-1· d^-1 for 2 days on the 6^th ,10^th ,13^th ,16^th day. The control group(Model) were only treated by BU 35mg ·kg^-1· d^-1 for 4 days and CY 50mg ·kg^-1· d^-1 for 2 days. Results:After BU used, the white blood cell of all the groups began to decline from the 3th day except Group6 was from the 5^th day. The minimum of white blood cell of all the groups ( Model, Group6, Group10 , Group13 , Group16 ) appeared on the 11^th, 9^th, 9^th, 11^th ,11^th day,and then restored to the normal level on the 19^th ,23^th ,23^th ,21^th and 19^th day. On the 19^th day after BU used, the bone marrow features were generally normal for Model and Group16 , and the myeloproliferative were poorer for the other groups. Conclusion:There was no influence for the recovery of bone marrow of the mice, treated by BU 35mg ·kg^-1· d^-1 for 4 days and CY 50mg ·kg^-1· d^-1for 2 days on the 16th day after injected subcutaneously by PEG - rhG - CSF. So we can prevent the decline of white blood cells by PEG - rhG - CSF in the chemotherapy which treatment time last 5 days per 3 weeks for clinical application.
