摘要
目的通过Meta分析评估血管紧张素转化酶(ACE)基因I/D多态性与中国人群消化道肿瘤发病的相关性。方法通过计算机文献检索中国生物医学文献数据库(CBM)和Pub Med数据库,并结合文献追溯的方法,收集所有关于ACE基因多态性与消化道肿瘤易感性的病例对照研究或队列研究。以消化道肿瘤组与对照组人群基因型分布的OR值为效应指标,各资料间进行一致性检验,以确定采用固定或随机效应模型进行合并分析。发表偏倚用漏斗图和Egger线性回归检验来评估。结果纳入在国内外发表的相关文献6篇,Meta分析结果表明:D等位基因相对于I等位基因合并OR值(95%CI)为1.32(0.46~3.79)(P〉0.05),DD基因型相对于DI/II基因型合并OR值(95%CI)为0.89(0.39~2.01)(P〉0.05),DD/DI基因型相对于Ⅱ基因型合并OR值(95%CI)为1.08(0.89~1.31)(P〉0.05),DD基因型相对于Ⅱ基因型合并OR值(95%CI)为0.86(0.43~1.75)(P〉0.05),DI基因型相对于II基因型合并OR值(95%CI)为1.02(0.74~1.40)(P〉0.05),差异均无统计学意义。结论中国人群ACE基因I/D多态性与中国人群消化道肿瘤易感性无明显关联。
Objective To study the relationship between angiotensin-converting enzyme( ACE) gene I/D polymorphisms and susceptibility to digestive system cancer in Chinese populations by Meta-analysis. Methods Computer literature searching was carried out in CBM and Pub Med database to collect the articles of the case-control studies on association between ACE polymorphism and susceptibility to digestive system cancer. Odds ratio was employed to evaluate the susceptibility to digestive system cancer and ACE status. Heterogeneity was checked by chi-square test. Publication bias was assessed by a funnel plot and Egger's test. Results Six papers were selected. Meta-analysis found that OR( 95% CI) of D allele compared with that of I allele was 1. 32( 0. 46 ~ 3. 79)( P 0. 05). OR( 95% CI) of DD genotype compared with that of DI / II genotype was 0. 89( 0. 39 ~ 2. 01)( P 0. 05). OR( 95% CI) of DD / DI genotype compared with that of II genotype was 1. 08( 0. 89 ~ 1. 31)( P 0. 05). OR( 95% CI) of DD genotype compared with that of II genotype was 0. 86( 0. 43 ~ 1. 75)( P 0. 05).OR( 95% CI) of DI genotype compared with that of II genotype was 1. 02( 0. 74 ~ 1. 40)( P 0. 05). Conclusions Angiotensin-converting enzyme polymorphisms may not be significantly associated with digestive system cancer in Chinese populations.
