摘要
盐酸土霉素是一种广谱抑菌剂,常用作兽用抗生素。现行工艺是在以土霉素碱为原料在甲醇中冷却结晶制得,但是得到的盐酸土霉素晶体具有晶粒小,形状不规则,流动性差等缺点,为了改进结晶工艺,本文测定了在不同温度下,盐酸土霉素在各种溶剂中的溶解度,并用半经验Apelblat方程和多项式经验方程进行拟合回归,拟合结果良好。试验中发现,盐酸土霉素澄清饱和溶液在搅拌过程中会变成乳白色液体,针对此异常现象的研究分析表明温度升高时能加剧一些副反应,如差向化反应,导致成品中的杂质含量超标。
Oxytetracycline hydrochloride is an antibiotic, widely used as pharmaceuticals for animals. Presently, oxytetracycline hydrochloride is obtained by cooling crystallization in methanol, with many disadvantages, including small crystals, diversifying shapes and low liquidity. In this paper, in order to reform crystallization technology, the solubilities of oxytetracycline hydrochloride in many organic solvents was investigated and the solubility data was correlated well with an Abelblat equation or empirical polynomial equation. Some abnormal phenomenon was also investigated, and the experiment result shows that the temperature increase of dissolving would lead to structural change and result in the impurities increase in the finished oxytetracycline product.
