摘要
目的通过超声引导建立兔肢体软组织肿瘤模型,为后期的干预研究和影像学评价奠定基础。方法选用雄性新西兰兔30只,6~8个月龄,体重3.5~4.0kg,随机分为对照组6只,模型组24只。两组应用磁共振弥散成像(MRDWI)和i维超声观察肿瘤的形态、信号特点及体积变化。对照组在采集影像后处死兔观察组织学变化。模型组于第14、21、28天随机分批处死6只兔观察组织学变化。剩下6只兔不处死,观察带瘤生存时间。结果三维超声显示:对照组在注射部位未见任何同声。模型组在第24小时、7、14、21、28天的肿瘤体积分别为:(1.61±0.18)、(4.15±0.27)、(7.92±0.50)、(15.02±0.70)、(18.23±0.67)cm^3。与对照组比较其差异有统计学意义(P〈0.05)。MRDWI:对照组未见明显的异常信号。模型组在左侧后肢可见肿瘤影像,肿瘤组织在MRDWI图呈高信号;在表观扩散系数(ADC)图呈低信号。组织学结果:对照组可址正常的横纹肌结构。模型组第21天大体标本观察,可见肿瘤实质呈现不规则的坏死区。第60天,解剖胸腔,可见两肺弥漫性转移结节。结论通过超声引导注射肿瘤组织悬液,成瘤率高,肿瘤生长范围较局限。影像学和组织学结果提示:后期的十预研究应在种植肿瘤后的第14天左右进行,以降低肿瘤实质液化坏死对实验结果的影响。
Objective Establish the rabbit limb soft tissue tumor model with ultrasound guidance for further intervention study and imaging evaluation. Methods 30 male New Zealand white rabbits were used and randomly divided into control group (n = 6 )and model group (n = 24). Magnetic resonance diffu- sion weighted imaging (MRDWI) and three - dimensional ultrasound were used in two groups to analyze tumor morphology, signal characteristics and volume changes. Histological changes were collected from sacrificed animals after capturing image in the control group. Six animals were randomly sacrificed at 14, 21 and 28 d after histological changes were captured in the model group. The remaining six animals were not killed, to observe survival time with tumor. Results Three - dimensional ultrasound findings showed that there was no abnormal echo at injection site in the control group. The tumor volume in the model group at24 hours, 7 days, 14 days, 21 days, 28 days was(1.06±0.15), (2.21 ±0.44), (3.38±1.23), and (4. 60 ± 0. 86 ) cm^3 , respectively, and the statistically was significant compared with the control group (P 〈0. 05). MRDWI showed that no abnormal signal was observed in the control groups. Tumor imaging was captured in the left hind limb, and high signal was detected in tumor tissues with DWI maps in model group, while low signal was observed with apparent diffusion coefficient (ADC) maps. Histology result: with hematoxylin and eosin (HE) staining, normal striated muscle was shown in the control group. In the model group with gross examination, revealed the irregular necrosis in the tumor parenchyma at the 21 days; Gross anatomy showed diffuse metastatic nodules on the lungs at the 60 d. Conclusion Inject tumor tissues suspension according to ultrasound guidance with a high tumor formation rate and limited tumor growth range.
