摘要
【目的】探究黄粉虫抗菌肽对耐药粪肠球菌抑菌效果。【方法】用浓度为1×108CFU/m L的粪肠球菌菌液与麸皮混合饲喂诱导黄粉虫幼虫,在24 h后提取黄粉虫抗菌肽,用考马斯亮蓝法(Bradford法)测定黄粉虫抗菌肽粗提液浓度,采用试管稀释法测定青霉素、庆大霉素对粪肠球菌的最小抑菌浓度(MIC),用多步诱导法筛选出青霉素和庆大霉素耐药的粪肠球菌,用Kirby Bauer法测定黄粉虫抗菌肽对耐青霉素和庆大霉素的粪肠球菌的抑菌活性。【结果】经粪肠球菌诱导24 h后提取的黄粉虫抗菌肽浓度,显著高于未用粪肠球菌诱导的对照组(P<0.05),其抑菌活性也显著高于对照组(P<0.05),并且诱导黄粉虫抗菌肽组对耐青霉素、庆大霉素粪肠球菌的抑菌效果显著高于抗生素组(P<0.05)。【结论】黄粉虫抗菌肽对耐药粪肠球菌具有显著的抑菌效果。
【Objective】In order to explore tenebrio molitor antimicrobial peptides( AMPs) and observe it for resistant Enterococcus faecalis inhibitory effect. 【Method】In this experiment,tenebrio molitor larva were fed with concentration of Enterococcus faecalis bacteria 1 × 108 CFU / m L mixed with bran,the AMPs was extracted crudely after 24 h respectively. Crude extract of the antibacterial peptides of tenebrio molitors was detected by Coomassie brilliant blue method. MIC was determined for enterococcus against penicillin and gentamicin by tube broth method. At last enterococcus were screened with multi- step inducing method and the antibacterial activity of peptides were determined by Kirby Bauer method. 【Result】The result showed that the value of peptides was obviously higher than these not induced by enterococcus in the control group( P〈0. 05). The antibacterial activity of peptides was also higher than that of the control group( P〈0. 05) and antibiotic group( P〈0. 05).【Conclusion】Antibacterial peptides of tenebrio molitors have a good effect on gentamicin resistant enterococcus.
