摘要 目的观察15-脂氧合酶-1(15-LOX-1)在结肠正常黏膜、增生性息肉、管状腺瘤、绒毛状腺瘤及腺癌中的表达,探讨15-LOX-1在结肠息肉恶变中的作用。方法选取肠镜检查的正常结肠黏膜15例,肠镜下摘除的增生性息肉25例、管状腺瘤38例、绒毛状腺瘤30例及活检结肠腺癌15例,采用RT-PCR检测各组织中15-LOX-1 mRNA的表达,免疫组织化学和Western blot法检测各组织中15-LOX-1蛋白的表达。结果RT-PCR结果显示,15-LOX-1 mRNA在结肠正常黏膜、增生性息肉、管状腺瘤、绒毛状腺瘤及腺癌组织中的表达逐渐降低,除管状腺瘤与绒毛状腺瘤组织之间差异无统计学意义(P=0.168)外,其余各组织之间差异均有统计学意义(P<0.05);免疫组织化学结果显示,15-LOX-1蛋白在结肠正常黏膜组织中表达最高,在腺癌组织中表达最低,除增生性息肉与正常黏膜组织之间差异无统计学意义(P=0.075),管状腺瘤与绒毛状腺瘤组织之间差异亦无统计学意义(P=0.125)外,其余各组织之间差异均有统计学意义(P<0.05);Western blot结果显示,15-LOX-1蛋白在结肠正常黏膜组织中表达最高,在腺癌组织中表达最低,除增生性息肉与正常黏膜组织之间差异无统计学意义(P=0.178)外,其余各组织之间差异均有统计学意义(P<0.05)。结论 15-LOX-1具有抑癌作用,其表达的降低或缺失可能促使结肠息肉发生恶变。 Objective To observe the expression of 15- lipoxygenase- 1( 15- LOX- 1) in normal colon mucosa,hyperplastic polyps,tubular adenoma,villous adenoma and adenocarcinoma of colon,and to investigate the effect of15- LOX- 1 on malignant transformation of colon polyps. Methods Fifteen cases of normal colon mucosa,25 cases of hyperplastic polyps,38 cases of tubular adenoma,30 cases of villous adenoma and 15 cases of adenocarcinoma of colon removed by enteroscopy were selected. 15- LOX- 1 mRNA expression in all tissues was detected by quantitative RT-PCR,and protein expression by immunohistochemistry and Western blot. Results RT- PCR showed that 15- LOX- 1mRNA expression in normal colon mucosa,hyperplastic polyps,tubular adenoma,villous adenoma and adenocarcinoma of colon was reduced gradually. There was no significant difference between tubular adenoma tissues and villous adenoma tissues( P = 0. 168). There were significant differences among all other tissues( P 0. 05). Immunohistochemical detection showed that 15- LOX- 1 protein expression was highest in normal colon mucosa tissues,yet lowest in adenocarcinoma of colon. There was no significant difference in 15- LOX- 1 protein expression between hyperplastic polyp tissues and normal colon tissues( P = 0. 075),as well as between tubular adenoma tissues and villous adenoma tissues( P = 0. 125).There were significant differences among all other tissues( P 0. 05). Western blot showed that 15- LOX- 1 protein expression was highest in normal colon mucosa,yet lowest in adenocarcinoma of colon. There was no significant difference between hyperplastic polyp tissues and normal colon mucosa tissues( P = 0. 178). There were significant differences among all other tissues( P 0. 05). Conclusions 15- LOX- 1 has an effect against carcinogenesis. Reduction or absence of its expression may facilitate malignant transformation of colon polyps.
