摘要
链格孢菌为空心莲子草最具开发潜力的生防菌之一。通过室内生物测定的方法比较了链格孢菌不同菌株对空心莲子草的致病性,并测定了培养基、温度及pH值条件对强致病菌株菌落生长和产孢的影响,优化了其培养条件。结果表明,菌株J-14的致病性最强,其次为菌株MS613、ZL500和PEC,而菌株ML03的致病性最弱,其所产生的病斑直径仅为J-14的22.12%。菌株J-14在空心莲子草茎叶煎汁培养基(ADA)上菌落生长和产孢量较高,培养5 d后其菌落直径和产孢量分别为5.800 cm和519.374×10^6个;温度20~30℃及p H值6.5~8.5是其菌落生长和产孢的适宜温度和酸碱度,且最适温度和p H值分别为25℃和7.5。由此可见,菌株J-14对空心莲子草具有较强的致病性,ADA培养基、25℃和pH值7.5是其最佳培养条件,为将J-14菌株开发为空心莲子草生防菌提供了重要理论依据。
Alternarla alternata is one of biocontroling agents against Alternanthera philoxeroides. Pathogenicity of different A. alternata strains against A. philoxeroides was investigated by bioassay in laboratory, and the effects of different culture mediums, temperature and pH on colo- ny growth and spore production of A. alternata were also determined to optimize the culture conditions. The results showed that: The patho- genicity of J-14 was the highest, followed by MS613, ZLS00 and PEC stxains, and the lowest for ML03 with 22.12 % necrosis diameter as J-14. J-14 strain could well grow and produce spores on ADA medium, the colony diameter and spore production were 5. 800 cm and 519. 374 × 10^6 , respectively; 20 -30℃ and pH 6.5 - 8.5 were the proper temperature and pH value, with the best optimal 25 ℃ and pH 7. 5. It was concluded that J-14 strain was the most pathogenic to A. philoxeroides, ADA medium, 25 ℃ and pH 7.5 were its optimal culture conditions, which provided important theoretical basis for developing it as biocontroling agent againt A. philoxeroides.
