摘要
壳聚糖是甲壳素部分脱乙酰的产物,天然存在于一些真菌细胞壁中,具有良好的生物相容性和生物可降解性,被广泛用于食品、医药等领域。采用稀硫酸法、碱-酸法、碱-酸结合α-淀粉酶法对雅致放射毛霉壳聚糖提取的影响,得出碱-酸结合α-淀粉酶法提取壳聚糖效果最佳。对碱-酸结合α-淀粉酶法进行条件优化,结果表明,最佳提取条件为:1mol/L Na OH在121℃处理30min,4%(v/v)盐酸在95℃处理6h,添加4%(v/v)的α-淀粉酶在90℃处理2h。采用该方法提取壳聚糖,壳聚糖得率可达16.91%,利用茜素红法测得壳聚糖纯度为92.83%。
Chitosan is partially deacetylated from chitin and exists in the cell wall of fungi.It has excellent biocompatibility and biodegradability and is widely used in foods and medicine.Chitosan extraction from Actinomucor elegans were compared by different extraction methods.Based on the purity and yield of chitosan,alkali and acid treatments followed by α-amylase treatment was chosen for further studies.The optimal conditions for chitosan extraction were :1M Na OH at 121℃ for 15 min and 4%(v/v)hydrochloric acid at 95℃ for 6 h and then adding 4%(v/v)α-amylase extracting at 90℃ for 2 h.The highest extraction yield of chitosan was 16.91%.Then alizarin red method was applied for purification.The results indicated 92.83% of purified chitosan was obtained.
