摘要
目的研究复方丹参滴丸对脂多糖(LPS)造抑郁样行为模型小鼠的抗抑郁作用。方法采用LPS注射复制小鼠抑郁样行为模型,分别观察复方丹参滴丸(FDD)高剂量(270μg/g)和低剂量(135μg/g)对抑郁模型小鼠旷场活动、强迫游泳不动时间的影响;并通过检测血清和大脑皮层的白细胞介素-6(IL-6)、肿瘤坏死因子-α(TNF-α)和白细胞介素-10(IL-10),从复方丹参滴丸对抑郁症模型炎性细胞因子作用角度解释其抗抑郁机制。结果 FDD高剂量组能显著提高小鼠旷场活动距离,FDD高、低剂量组都能显著提高小鼠站立次数,并显著减少强迫游泳不动时间,差异有统计学意义(P<0.05);血清中,FDD各剂量组都能够显著降低IL-6、TNF-α水平,升高IL-10水平,差异有统计学意义(P<0.05,P<0.01);大脑皮层中,FDD各剂量组(P<0.01)都能够显著对抗LPS引起的IL-6升高,FDD高剂量组都能够明显对抗LPS引起的TNF-α升高(P<0.05),FDD高剂量组(P<0.01)、低剂量组(P<0.05)都能够显著对抗LPS引起的IL-10升高。结论 FDD对LPS造抑郁样行为模型小鼠抑郁症状的改善可能与其抗炎免疫作用有关,具体机制有待进一步研究。
Objective To study antidepressant effects of "Compound Danshen Dripping Pill "( FDD) on lipopolysaccharide( LPS)-induced depressive-like behaviors in mice. Methods To observe the influence of FDD high dosage( 270 μg / g) and low dosage( 135 μg / g) on open field test( OFT) and forced swim test( FST) in depressive-like behavior model mice induced by LPS injection,furthermore,to detect interleukin 6( IL-6),tumor necrosis factor alpha( TNFalpha) and interleukin 10( IL-10) levels in serum and cerebral cortex,explain antidepressant mechanism of FDD from its effects on inflammatory cytokines in depression model. Results High dosage of FDD can obviously improve the decrease of moving distances in OFT induced by LPS injection,and both of high and low dosage of FDD can remarkable increase the decreasing of rearing times induced by LPS,as well as decrease the improvement of immobility time of mice in FST induced by LPS. All of these results are statistically significant( P〈0.05). In serum,each dosage of FDD can significantly reduce IL-6,TNF-α level improvement,and increase IL-10 decrease induced by LPS with statistically significant( P〈0.05,P〈0.01). In cerebral cortex,each dosage of FDD can significantly reduce IL-6 level improvement induced by LPS( P〈0.01),high dosage of FDD can obviously reverse the improvement of TNF-α level induced by LPS( P〈0.05),and both of high dosage( P〈0.01) and low dosage( P〈0.05) can obviously reverse IL-10 level increase. Conclusion The improvement effects of FDD on depressive-like behaviors mice model induced by LPS may be related to its anti-inflammation and immune system regulation effects,and its mechanism needs further research.
