摘要
[目的]研究永福杜鹃组织培养离体再生体系。[方法]以永福杜鹃的带腋芽幼嫩茎段、带顶芽茎尖和不带顶芽茎尖为外植体,采用不同激素配比进行组培快繁研究,建立永福杜鹃离体快繁技术体系。[结果]适宜永福杜鹃离体培养的外植体为带顶芽茎尖,外植体消毒以0.1%氯化汞消毒5 min为宜,芽诱导最适培养基为1/2MS+2-ip 3.0 mg/L+NAA 0.10 mg/L+活性炭0.5 g/L,生根最适培养基为1/2MS+NAA 0.8 mg/L。[结论]建立了永福杜鹃组织培养离体再生体系,为永福杜鹃离体快繁技术研究提供理论依据。
[ Objective J The a^m was to study m vxtro regeneration culture system ot ~ ongtu azalea. L lwetnou 3 lne nssue raptu propagauon was conducted on stem segments with axillary buds, stem apex with top buds, and stem shoot without top buds as explants, using various hor- mone ratios, in vitro rapid propagation technique system was established. [ Result] Using stem apex as explant was appropriate for in vitro cul- ture of azalea, sterilization of explants with 0. 1% mercuric chloride for 5 minutes was suitable, the optimum bud induction medium was 1/2MS + 2-ip 3.0 mg/L + NAA 0.10 mg/L + activated carbon 0.5 g/L, the optimum rooting medium was 1/2MS + NAA 0.8 mg/L. [ Con- clusion] The in vitro regeneration culture system for Yongfu azalea is established, which can provide theoretical basis for research of Yongfu a- zalea in vitro rapid propagation technology.
