摘要
目的探讨幽门螺杆菌(helicobacter pylori,Hp)在胃癌组织中的感染情况及其与ABCE1基因的相关性。方法检测44例胃癌和38例癌旁正常胃组织标本Hp感染情况;采用反转录聚合酶链反应(RT-PCR)和蛋白印记(Western blot)技术检测Hp阳性、阴性胃组织中ABCE1 mRNA、ABCE1蛋白表达情况。结果胃癌组织中Hp感染率显著高于癌旁正常胃组织(P<0.05);低分化胃癌组织Hp感染率显著高于中-高分化胃癌组织(P<0.05);Hp阳性胃组织中ABCE1 mRNA及ABCE1蛋白表达均显著高于Hp阴性胃组织(P<0.01);低分化胃癌组织ABCE1 mRNA及ABCE1蛋白表达均显著高于中-高分化胃癌组织(P<0.05)。结论胃癌的发生、发展与Hp感染及ABCE1基因表达有关,临床可通过Hp和ABCE1基因检测筛查早期胃癌,及时治疗,以降低病死率、改善患者生存质量。
Objective To investigate the infection status of helicohacter pylori (Hp) in gastric cancer tissues and its correlation with ABCE1 gene. Methods Hp infection status of 44 cases of gastric cancer tissues and 38 cases of normal gastric tissues of adjacent cancer tissues were detected ; ABCE1 mRNA and protein expressions in Hp positive and negative gastric tissues were detected using reverse transcriptionpolymerase chain reaction (RT-PCR) and Western blot methods. Results Hp infection rate in gastric cancer tissues was significantly higher than that in normal gastric tissues of adjacent cancer tissues ( P 〈 0. 05 ) ; Hp infection rate in poorly differentiated gastric cancer tissues was significantly higher than that in middle-highly differentiated gastric cancer tissues (P 〈 0. 05) ; the values of ABCE1 mRNA and protein expressions in Hp positive gastric tissues were significantly higher than those in Hp negative gastric tissues ( P 〈 0. 01 ) ; the values of ABCE1 mRNA and protein expressions in poorly differentiated gastric cancer tissues were significantly higher than those in middle-highly differentiated gastric cancer tissues (P 〈 0. 05). Conclusion The occurrence and development of gastric cancer is related to Hp infection and ABCE1 gene expression, and therefore clinicians may find the early gastric cancer by detecting the Hp snf ABCE1 gene so as and treat timely and reduce the fatality rate of gastric cancer and improve the patients'quality of life.
