摘要
目的:探讨健脾方对动脉粥样硬化(AS)Apo E基因敲除(Apo E-/-)小鼠心脏和血清5-羟色胺(5-HT)和5-羟吲哚乙酸(5-HIAA)表达的影响,进而明确其作用机制。方法:将Apo E-/-小鼠随机分为模型组,西药对照组(阿托伐他汀),健脾方低、中、高剂量组,饲以高脂饲料,每组10只;将同系C57BL/6小鼠随机分为空白对照组,饲以普通饲料,单纯药物干预组(健脾方中剂量),饲以高脂饲料,每组10只。空白对照组和模型组予0.9%氯化钠溶液灌胃,其余组给予同等剂量的对应药物灌胃,12周灌胃结束后,各组采用ELISA法测定心脏和血清5-HT、5-HIAA含量。结果:与空白对照组比较,模型组5-HT、5-HIAA含量显著升高(P<0.01),5-HT的转化率显著下降(P<0.01);而与模型组比较,各治疗组5-HT和5-HIAA水平显著下降(P<0.01),5-HT转化率也显著降低(P<0.01)。结论:Apo E-/-小鼠早期AS的形成可能与5-HT的代谢减弱使心脏与血清中5-HT高表达有关;健脾方干预早期AS的机制可能与降低心脏和血清中5-HT、5-HIAA的含量有关。
Objective:To investigate the effect of Jianpi Formula on expression of 5-HT and 5-HIAA in heart and serum of ApoE-/- mice model with early atherosclerosis(AS),in order to clarify its mechanism.Methods:The ApoE-/- mice were randomly divided into model group,western medicine control group(atorvastatin group),low,middle and high dosage of Jianpi Formula groups,and with 10 mice in each group.The homologous C57BL/6 mice were randomly divided into blank group and pure drug intervention group,and with 10 mice in each group.Mice in blank control group were fed with normal diets,and mice in the other groups were fed with high-fat diet.Mice in blank control group and model group received gavage administration with normal saline,and mice in the other groups received gavage administration with corresponding drugs for 12 weeks.The contents of 5-HT and 5-HIAA in serum and heart tissues of mice were tested by using ELISA.Results:Compared with blank control group,the contents of 5-HT and 5-HIAA of mice in model group increased significantly(P〈0.01),and the conversion rate of 5-HT decreased significantly(P〈0.01).Compared with model group,the contents of 5-HT and 5-HIAA and conversion rate of 5-HT of mice in treatment groups decreased remarkably(P〈0.01).Conclusion:The early formation of AS in ApoE-/- mice might be related with the high expression of 5-HT in serum and heart tissues.The intervention mechanism of Jianpi Formula on early AS might be related with the decreasing of contents of 5-HT and 5-HIAA in serum and heart tissues.
