摘要
目的:研究胡桃楸茎枝含药鸡蛋中吸收成分的提取工艺,为胡桃楸的抗肿瘤有效成分研究奠定基础。方法:采用不同溶剂提取胡桃楸茎枝的含药鸡蛋提取液作为供试品溶液,以供试品溶液中HPLC峰个数及峰面积总和为评价指标。通过单因素试验对提取溶剂中加酸量、提取方法和提取溶剂种类进行选择,通过正交试验对提取溶剂的浓度、提取溶剂的用量和提取时间进行优化。结果:通过单因素试验确定提取溶剂中不加酸,采用回流提取,提取溶剂为含水丙酮;正交试验确定最佳提取工艺为:50倍量50%丙酮回流提取2h。结论:优选后的提取工艺稳定可靠、合理可行,为研究胡桃楸含药鸡蛋中吸收的成分奠定了基础。
Objective Eggs cooked with the branches of Juglans mandshurica which was called drug - contained eggs. For the re- search of constituents absorbed by the drug - contained eggs, this study was conducted to optimize the extraction technology of the drug - contained eggs. Methods For the optimization of acid content added into the extracting solvent, extracting method, and extracting solvent, single factor experiment was designed. Orthogonal experiment was conducted to choose the time of extraction, concentration and volume of the extracting solvent. The total number and the sum of area of chromatographic peak were taken as investigation index. Results The result of single factor experiment demonstrated that acetone and reflux was suitable for the extraction and no acid should be added into the extracting solvent. Orthogonal experiment suggested that the drug - contained eggs should be extracted with 50 times of 50% acetone by reflux for 2 hours. Conclusions The optimum extracting technology is reasonable and stable. This work was useful for the research of the constituents absorbed by the drug - contained eggs.
