摘要
目的研究淡豆豉(Semen Sojae Praeparatum)的化学成分及其促进大鼠颅骨细胞增殖的活性。方法采用反复硅胶柱色谱、MCI和Sephadex LH-20柱色谱等方法进行分离纯化,并通过化合物的理化性质和波谱数据进行结构鉴定。采用MTT法考察化合物对大鼠颅骨细胞增殖的作用。结果分离得到12个化合物,其中7个黄酮化合物:大豆苷(1),大豆苷元(2),染料木素(3),染料木苷(4),黄豆黄素(5),黄豆黄苷(6),芹菜素(7);3个甾类化合物:β-谷甾醇(8),豆甾醇(9),菜油甾醇(10);2个小分子芳香化合物:丁香醛(11),丁香酸(12)。结论化合物5~11均为首次从淡豆豉中分离得到。5个异黄酮类化合物具有一定的促成骨细胞增殖活性,且在1×10^(-9)~1×10^(-6)mol·L^(-1)内细胞增殖呈现明显的剂量依赖,3个黄酮苷元(大豆苷元、染料木素、黄豆黄素)的促成骨细胞增殖活性优于2个黄酮苷(大豆苷、染料木苷)。
OBJECTIVE To study the chemical constituents and their proliferation activity on rat osteoblasts of Semen Sojae Pra- eparatum. METHODS The compounds were isolated by chromatography on silica gel column, MCI and sephadex LH -20 column. Their structures were elucidated by spectral analysis. The proliferation test was performed by using rat osteohlasts with MTT assays in vitro. RESULTS Twelve compounds were obtained and identified as daidzin ( 1 ), daidzein ( 2 ), genistein ( 3 ), genistin ( 4 ), glycitein ( 5 ), glycitin( 6 ), apigenin( 7 ), ^-sitosterol ( 8 ), stigmasterol ( 9 ), eampesterol ( 10 ), syringaldehyde ( 11 ), syringie acid ( 12 ), respectively. CONCLUSION Compounds 5 - 11 are isolated from Semen Sojae Praeparatum for the first time. The five isoflavones show proliferation activity on rat osteoblasts, and aglycones proved to show better activity than glycosides.
