摘要
目的探讨葡萄酒以及同样含有酚类的葡萄汁是否对急性酒精性肝损伤具有保护作用。方法小鼠随机分为4组:对照组、酒精组、葡萄酒组和葡萄汁组,后三组每天灌胃0.4 ml30%的食用酒精,连续7 d,建立急性酒精性肝损伤模型。同时,葡萄酒组灌胃0.42 ml长城干红葡萄酒(12%vol),葡萄汁组灌胃葡萄汁(原果汁含量100%)0.42 ml,对照组与酒精组灌入0.42 ml的蒸馏水。称量实验前后小鼠体重,检测实验后血清谷丙转氨酶(ALT)活性和丙二醛(MDA)含量,并对肝脏做病理检查。结果对照组和葡萄汁组实验前后体重增加有统计学意义(P<0.01),酒精组体重变化无统计学意义,葡萄酒组体重下降有统计学意义(P<0.05);葡萄酒组与对照组、葡萄汁组相比,ALT活力增加有统计学意义(P<0.05),而葡萄汁组与对照组、酒精组的差异均无统计学意义;各组间的MDA差异均无统计学意义;病理切片结果显示,葡萄汁与对照组肝小叶结构清晰,着色良好,细胞索排列整齐,而酒精和葡萄酒组肝小叶界限不清,排列紊乱,在中央静脉周围出现大量脂肪空泡。结论葡萄酒对急性酒精性肝损伤小鼠有损害作用,葡萄汁具有保护作用。
Objective To investigate whether wines and phenolic containing grape juice has a protective effect on acute alcoholic liver injury. Methods Mice were divided into four groups: normal control,alcohol,wine,and grape juice. All mice except the control group were under intragastric administration of 0.4 ml 30 % alcohol to establish alcoholic liver injury model. The wine group was under intragastric administration of 0.42 ml Great Wall wine(12 %vol). The grape juice group was under intragastric administration of Malee grape juice. Administration continued for 7 days. The weight,serum alanine transaminase(ALT) and malondialdehyde(MDA) of the mice were measured. Pathological sections were made from mice livers.Result Control group and grape juice group weight significantly increased(P0.01),and alcohol group weight did not change significantly,but wine group weight decreased significantly(P0.05). In the wine group,ALT activity was significantly higher than those of control and grape juice group(P0.05). ALT activity of the grape juice group was not significantly different from that of control and alcohol group. Pathological section showed that the structures of hepatic lobule of control and grape juice group were very clear and the cells were neatly arranged. On the other hand,the structures of hepatic lobule of the wine and alcohol group were disorganized and a large number of fat vacuoles around the central vein was observed. Conclusion Wine has an adverse effect on alcoholic liver injury, but grape juice can protect it.
