摘要
目的探究湖南本土种植及加工的鸡头黄精、多花黄精采用不同炮制方法对黄精多糖含量的影响。方法超声提取法提取多糖,用Sevage法除去蛋白,采用苯酚-硫酸法测定多糖含量。结果鸡头黄精经二蒸二晒、四蒸四晒、七蒸七晒、九蒸九晒炮制后,多糖含量分别为7.973%、3.584%、2.845%、2.194%;多花黄精经二蒸二晒、四蒸四晒、五蒸五晒、七蒸七晒炮制后,多糖含量分别为9.602%、3.185%、2.572%、2.043%;精密度良好(RSD=0.274%)、平均回收率99.29%、试验在60 min内稳定性良好(RSD=1.36%)。结论炮制过程蒸晒次数越多,黄精多糖含量越少。
Objective To explore different processing methods on polysaccharide content of Polygonatum sibiricum Red.and Polygonatum cyrtonema Hua, which cultivated and processed in Hunan. Methods The polysaccharide was extracted by ultrasonic extraction and the protein was removed by Sevage’s method. The content of polysaccharide was determined by phenol-sulfuric acid method. Results The polysaccharide content of Polygonatum sibiricum Red. with different times of steaming and drying processing methods were 7.973%, 3.584%, 2.845%, 2.194%, and the polysaccharide content of Polygonatum cyrtonema Hua with different processing methods were 9.602%, 3.185%, 2.572%, 2.043%; The precision was good(RSD =0.274%); The average recovery rate was 99.29%; the test has good stability in 60min(RSD=1.36%). Conclusion The content of polysaccharide was reduced with the more times of steaming and drying processing methods.
