摘要
目的观察硒对肺腺癌A549细胞增殖和凋亡的影响,探讨硒诱导肺癌细胞凋亡的机制。方法设不同浓度硒(1,5,10μmol/L)处理实验组和一个阴性空白对照组,采用MTT法检测细胞抑制率;FCM仪分析细胞的凋亡率;Western blot法检测凋亡相关分子Bcl-2,Bcl-x L,Bax,细胞色素C(cytochrome C)及caspase-3表达变化。结果与24 h组比较,同浓度(5μmol/L和10μmol/L)硒处理48 h组和72 h组显著抑制细胞增殖(P<0.05),且具有时间依赖性;与同时间对照组比较,硒能够明显抑制细胞增殖(P<0.05),且具有浓度依赖性。与对照组比较,硒能够显著性诱导细胞凋亡(P<0.05),降低Bcl-2,Bcl-x L的表达并增加Bax,cytochrome C和caspase-3的表达(P<0.05),且均具有浓度依赖性。结论硒通过调控Bcl-2,Bax,细胞色素C及caspase-3的表达诱导A549细胞凋亡。
Objective To investigate the effects of selenium on proliferation and apoptosis of lung adenocarcinoma cell line A549 cells and explore its mechanisms. Methods The A549 cells were treated with different concentrations of selenium( 1,5,10 μmol / L) for24 h and 48 h respectively,and the untreated cells were used as negative contols. The cell proliferation was measured using MTT,the cell apoptosis was measured by flow cytometry and the expression of Bcl-2,Bcl-xL,Bax,cytochrome C and caspase-3 was examined by Western blot. Results Compared with 24 h group,the proliferation was significantly inhibited after treated with the same concentration of selenium( 5,10 mol / L) in 48 h group and 72 h group( P〈0. 05). Selenium significantly inhibited the proliferation in a concentration-dependent manner at the same time( P〈0. 05). Selenium significantly induced the apoptosis( P〈0. 05),markedly down-regulated the expression of Bcl-2 and Bcl-xL and up-regulated expression of Bax,Cytochrome C and Caspase-3 in a concentration-dependent manner( P〈0. 05). Conclusion Selenium can induce the apoptosis of A549 cells by regulating the expression of Bcl-2,Bax,cytochrome C and caspase-3.
