摘要
目的:研究加味麻黄蝉衣汤对变应性鼻炎(AR)模型豚鼠的改善作用及其机制。方法:将32只豚鼠随机均分为正常(0.9%氯化钠注射液)组、模型(0.9%氯化钠注射液)组、实验[加味麻黄蝉衣汤,5.18 g(生药)/kg]组和对照[丙酸氟替卡松鼻喷雾剂,50μg/(只·d)]组,除正常组外其余各组豚鼠ip卵清蛋白和氢氧化铝各30 mg复制AR模型。除对照组喷雾给药外,其余各组ig相应药物,每天2次,连续15 d。观察豚鼠的鼻部症状并进行评分,免疫组化法检测鼻黏膜酪氨酸磷酸酶1(SHP-1)的蛋白表达水平,酶联免疫吸附法检测血清中白细胞介素4(IL-4)、干扰素γ(IFN-γ)的含量。结果:与正常组比较,模型组豚鼠出现AR症状,评分增加,鼻黏膜SHP-1蛋白表达水平升高,血清中IL-4含量增加、IFN-γ含量减少(P<0.05);与模型组比较,实验组和对照组豚鼠AR症状改善,评分降低,鼻黏膜SHP-1蛋白表达水平降低,血清中IL-4含量减少、IFN-γ含量增加(P<0.05)。结论:加味麻黄蝉衣汤对AR有一定的改善作用,其机制可能与减少血清中IL-4的含量、增加血清中IFN-γ的含量有关。
OBJECTIVE:To study improvement effect of modified mahuang chanyi decoction on allergic rhinitis(AR)model guinea pigs and its mechanism. METHODS:32 guinea pigs were randomly divided into normal group(0.9% Sodium chloride injection),model group(0.9% Sodium chloride injection),experimental [Modified mahuang chanyi decoction,5.18 g(medicinal materials)/kg] group and control [Fluticasone propionate nasal spray,50 μg/(guinea pig·d)] group. Except for normal group,other groups received ovalbumin 30 mg and calmogastrin 30 mg intraperitoneally to induce AR model. Except for control group,other groups were given relevant medicine intragastrically,twice a day for consecutive 15 d. Nasal symptoms of guinea pigs were observed and recorded,and then analyzed and scored. The protein expression of nasal mucosa SHP-1 was detected by immunohistochemistry,the serum contents of IL-4 and IFN-γ were determined by enzyme-linked immunosorbent assay. RESULTS:Compared with normal group,model group suffered from AR symptom,and nasal symptoms score was increased;and the protein expression of nasal mucosa SHP-1 and the serum content of IL-4 increased,while the content of IFN-γ decreased(P〈0.05). Compared with model group,AR symptom of experimental group and control group were improved;nasal symptoms score,the protein expression of nasal mucosa SHP-1 and the serum content of IL-4 decreased while the content of IFN-γ increased(P〈0.05). CONCLUSIONS:Modified mahuang chanyi decoction shows certain improvement effect on AR,and its mechanism is associated the decrease of IL-4content and the increase of IFN-γ content in serum.
