热应激对体外培养小鼠乳腺上皮细胞cAMP、SOD和MDA水平的影响 被引量:3

Effect of heat stress on the levels of cAMP,SOD,and MDA in mouse mammary epithelial cells cultured in vitro

摘要 为了研究在不同热应激强度处理下,小鼠乳腺上皮细胞环磷酸腺苷(c AMP)水平、超氧化物歧化酶(SOD)活性和丙二醛(MDA)含量的变化,试验将传至第3代的乳腺上皮细胞分为对照组(37℃)和热应激组,其中热应激组分为39℃组(0.5,1,1.5,2 h)和41℃组(0.5,1,1.5,2 h),同时提供5%CO2和饱和湿度的培养环境。结果表明:与37℃对照组相比,39℃培养1,1.5 h的小鼠乳腺上皮细胞c AMP水平显著升高(P<0.05);39℃培养2 h和41℃培养0.5,1,1.5,2 h的小鼠乳腺上皮细胞c AMP水平极显著升高(P<0.01)。与37℃对照组相比,39℃培养1.5 h的小鼠乳腺上皮细胞SOD活性显著高于对照组(P<0.05),39℃培养1 h、41℃培养0.5,1,1.5,2 h的小鼠乳腺上皮细胞SOD活性极显著高于对照组(P<0.01)。与37℃对照组相比,41℃培养1.5 h的小鼠乳腺上皮细胞MDA含量显著高于对照组(P<0.05),39℃培养0.5,1,1.5,2 h和41℃培养1 h的细胞内MDA含量极显著高于对照组(P<0.01)。说明随着热应激强度加强,小鼠乳腺上皮细胞的c AMP浓度上升;随着温度升高,MDA含量有下降趋势,SOD活性明显上升;在不同的热应激时间下,细胞的SOD活性呈下降趋势,MDA含量处于波动状态。 To study changes in the level of cyclic adenosine monophosphate( cAMP),superoxide dismutase( SOD) activity,and malondialdehyde( MDA) content in the mouse mammary epithelial cells under different intensity of heat stress,the third- generation mammary epithelial cells were divided into a control group( 37 ℃) and two heat stress groups,including 39 ℃( for 0. 5 h,1 h,1. 5 h,and 2 h) and 41 ℃( for0. 5 h,1 h,1. 5 h,and 2 h) with providing the training environment of 5% of CO2 and saturated humidity. The results showed that the levels of c AMP in the mouse mammary epithelial cells cultured at 39 ℃ for 1 h and 1. 5 h were significantly increased( P < 0. 05) compared with the control group; the levels of c AMP in the mouse mammary epithelial cells cultured at 39 ℃ for 2 h and at 41 ℃ for 0. 5 h,1 h,1. 5 h and 2 h were highly significantly increased( P < 0. 01) compared with the control group. The level of SOD activity in the mouse mammary epithelial cells cultured at 39 ℃ for 1. 5 h was significantly higher( P < 0. 05) than that in the control group,and the levels of SOD activity in the mouse mammary epithelial cells cultured at 39 ℃ for 1 h and at 41 ℃ for 0. 5 h,1 h,1. 5 h and 2 h were highly significantly higher( P < 0. 01) than those in the control group. The MDA content in the mouse mammary epithelial cells cultured at 41 ℃ for 1. 5 h was significantly higher( P <0. 05) than that in the control group,and the contents of MDA in the mouse mammary epithelial cells cultured at 39 ℃ for 0. 5 h,1 h,1. 5 h,and 2 h and at 41 ℃ for 1 h were highly significantly higher( P < 0. 01) than those in the control group. The results indicate that,the levels of c AMP are increased when strengthening of the intensity of heat stress,MDA content has a downward trend and the SOD activity is increased dramatically with the increase of temperature. Under different time of heat stress,the cellular SOD activity shows a downward trend,and the content of MDA is in a state of fluctuation.
