摘要
糖尿病与中枢神经系统性疾病密切相关,大量研究表明,血脑屏障功能障碍在糖尿病介导的中枢神经系统性疾病(如中风、血管性痴呆及记忆缺失等)中起着重要作用。机体血糖水平的改变(高血糖或低血糖)影响血脑屏障对葡萄糖、氨基酸等物质的转运,影响血脑屏障的完整性,而且与中枢神经系统微血管的氧化应激密切相关。但是,由于与糖尿病相关的中枢神经系统并发症的潜在发病因素比较复杂,因此发病机制方面还有待深入研究探讨。笔者综述近年来糖尿病对血脑屏障完整性及其功能影响的机制研究进展,以期为治疗糖尿病中枢神经系统并发症开拓新的研究方向,为开发治疗糖尿病中枢神经系统并发症提供新的药物作用靶点。
Diabetes mellitus is closely related to the central nervous system (CNS) diseases, and it is now evident that blood-brain barrier (BBB) dysfunction plays a significant role in diabetes-dependent CNS disorders, such as stroke, vascular dementia, cognitive deficits, etc. Changes in plasma glucose levels (hyper- or hypoglycemia) have been associated with altered BBB transport functions (e. g. , glucose, amino acids, etc. ) , the iutegrity of BBB, and oxidative stress in the CNS microvascular. However, the underlying causes of diabetes-induced CNS complications are multifactorial and are relatively little understood. This review focuses on the major findings and current knowledge with regard to the impact of diabetes on BBB integrity and function to explore the new research area and the new drug targets for treatment of diabetic dependent CNS complications.
