摘要
目的 了解MICA等位基因在中国北方汉族的分布及与HLA-B基因型的连锁关系。方法 用直接测序法对北方汉族224个标本的HLA-B和MICA基因的2—6外显子进行测序。用Arlequin软件统计分析2个位点间的连锁关系。结果 发现18个MICA的等位基因,其中MICA~*002(16.07%),MICA~*008(23.21%)和MICA~*010(21.65%)最为常见。MICA~*010-HLA-B~*46∶01,MICA~*045-HLA-B~*13∶01,MICA~*049-HLA-B~*51∶01有明显的连锁关系。结论 北方汉族人群呈现特异MICA等位基因库,MICA与HLA-B具有连锁关系。
Objective To study MICA polymorphism and MICA-HLA-B linkage disequilibrium in Northern Han population.Methods The exons 2 to 6 of HLA-B and MICA gene in 224 samples of northern Han population were sequenced by PCR sequence-based-typing method.The linkage disequilibrium between the two loci was statistically analyzed by Arlequin software.Results 18 alleles of MICA were detected in 224 samples.The frequency of MICA*002(16.07%),MICA*008(23.21%)and MICA*010(21.65%)were the most common in Northern Han population.There was linkage disequilibrium between MICA*010-HLA-B*46∶01,MICA*045-HLA-B*13∶01,MICA*049-HLA-B*51∶01.Conclusion There was a specific distribution of MICA allele in northern Han population,and there was a linkage disequilibrium between MICA and HLAB.
