沙棘叶中科罗索酸浓缩物的测定方法 被引量:4

摘要 对沙棘叶中科罗索酸的浓缩和提取方法进行了初步探究,提出了沙棘叶中科罗索酸含量的测定方法。结果显示,最佳提取方法是水-乙醇提取法,提取率为0.8%;提取物测定的色谱条件为:20RBAXSB-C_(18)(4.6 mm×250 mm,5μm)色谱柱,以甲醇-0.1%冰醋酸水溶液(体积比为88∶12)为流动相,检测波长为215 nm,流速为1 m L/min,柱温为30℃;科罗索酸在0.042 6~0.852 0 mg/m L范围内,峰面积与浓度有着良好的线性关系,平均回收率为105%,相对标准偏差(RSD)为1.75%(n=5)。该提取方法简便、安全、无有机物残留,适合于工业生产;测定方法准确、重复性好,可用于沙棘叶提取物的含量测定和质量控制。 The concentration and extraction method of corosolic acid in the seabuckthorn leaves was explored preliminarily and the assay method of corosolic acid content from the seabuckthorn leaves extractive was proposed in this paper. The results showed that the best extraction method was water-ethanol extraction method, the extraction rate of 0.8%. Chromatographic condition for the determination of extracts: 20RBAXSB-C_(18)(4.6 mm×250 mm,5 μm)column was adopted. The mobile phase was methanol-0.1% acetic acid aqueous solution(88 ∶12). The detection wavelength was set at 215 nm, flow rate was 1.0 m L/min, column temperature was 30 ℃. The concentration of corosolic acid was linear in the range of 0.042 6-0.852 0 mg/m L and the front area and concentration had good linear relationship. The average recovery rate was 105% and the RSD was 1.75%(n=5). The method was simple, safe, non-organic residue, it could be used for industrial production. Measuring method was accurate, reproducible, and it could be used for the determination and quality control of sea-buckthorn leaves extract.
