摘要
【目的】优选救心菜不定芽增殖培养基,为建立救心菜再生体系提供技术支持。【方法】以救心菜叶片为外植体采用L_9(3~4)正交试验设计,分析不同浓度6-BA、IBA、NAA和AgNO3组合对救心菜叶片愈伤组织诱导及不定芽增殖的影响。【结果】以75%乙醇(0.5 min)结合0.1%HgCl2(10.0 min)处理外植体表面的灭菌效果最佳;随着6-BA浓度的升高,愈伤组织的诱导效果越佳诱导率最高可达93.33%;各生长调节剂对救心菜不定芽分化和增殖影响排序为6-BA〉NAA〉IBA〉AgNO3,最适宜外植体芽诱导的培养基为6-BA 4.0 mg/L+NAA 0.3 mg/L+AgNO3 10.0 mg/L。经优选救心菜植株再生体系扩繁的组培苗生根率达100.00%,移栽成活率在98.00%以上。【结论】救心菜植株再生体系为:以75%乙醇处理0.5 min结合0.1%HgCl2处理10.0 min进行救心菜外植体表面灭菌,以MS+6-BA 4.0mg/L+NAA 0.3 mg/L+AgNO3 10.0 mg/L为外植体芽诱导最适宜培养基;利用该体系可进行救心菜组培苗扩繁。
[Objective]The multiplication medium optimization of adventitious buds for Sedum aizoon L.was optimized,in order to provide technical supports for establishing regeneration system of S.aizoon L..[Method]Using leaves of S.aizoon L.as explants,the L9(3~4) orthogonal experiment was conducted to investigate effects of different growth regulator combinations(6-BA,IBA,NAA and AgNO3) on callus induction and adventitious bud multiplication.[Result]The results showed that,when explants were treated with 75%alcohol for 0.5 min and 0.1%HgCl_2 for 10.0 min,the sterilization effect of this treatment method on explant surface was optimal.With the increase of concentration of 6-BA,the callus induction effect of 6-BA got better,the induction rate could reach up to 93.33%.Furthermore,the growth regulators affecting adventitious bud differentiation and proliferation of S.aizoon L.were ranked based on their influence extents,as follows:6-BANAAIBAAgNO3.And the optimal medium for bud induction of explants was MS+6-BA 4.0 mg/L+NAA 0.3mg/L+AgNO310.0 mg/L.Under optimized plant regeneration system,the rooting rate of tissue-cultured plantlets was found to be 100%and its transplanting survival rate was up to 98%.[Conclusion]The established plant regeneration system of S.aizoon L.is as follow:explants are treated with 75%alcohol for 0.5 min and 0.1%HgCl2 10.0 min to sterilize explant surface;the optimal medium for inducting adventitious bud is MS+6-BA 4.0 mg/L+NAA 0.3 mg/L+AgNO3 10.0 mg/L.Using above-mentioned system,the tissue-cultured plantlets of S.aizoon L.can be producted.
