摘要
目的探讨牡荆素对脂多糖诱导胰岛INS-1细胞损伤的保护作用。方法以脂多糖诱导INS-1细胞损伤,细胞分为对照组、模型组和牡荆素组(50μmol·L^(-1))。MTT法测定细胞存活率;TUNEL染色法检测细胞凋亡;RT-PCR法和Western blot法检测相关细胞凋亡因子Caspase-3、Bcl-2 mRNA和蛋白水平的表达。结果 5 mg·L^(-1)脂多糖可以诱导INS-1细胞凋亡,同时Bcl-2的表达明显减少,而Caspase-3的表达增加。经50μmol·L^(-1)牡荆素处理后,脂多糖诱导的INS-1细胞凋亡明显减少,Bcl-2的表达明显增加而Caspase-3的表达明显降低。结论牡荆素可抑制脂多糖诱导的INS-1细胞损伤及凋亡,其作用可能与调节Caspase-3、Bcl-2的表达有关。
Objective To study the protective effect of vitexin on lipopolysaccharides( LPS)-induced injury in INS-1 cells.Methods INS-1 cells apoptosis are induced by LPS,randomly divided into control,model and vitexin( 50 μmol·L^(-1)) groups. The cell viability of INS-1 was measured with MTT assay. Apoptosis rate of INS-1cells was detected with TUNEL staining. The expression of Caspase-3and Bcl-2 mRNA were determined by RT-PCR. The protein levels of Caspase-3,and Bcl-2 were determined by Western blot. Results The5 mg ·L^(-1)LPS could significantly induced injury of INS-1 cell line,leading to apoptosis rate of INS-1 cells increasing. Meanwhile,LPS could decrease the expression of Bcl-2,but the expression of Caspase-3 increased. In the presence of 50 μmol · L^(-1)vitexin,LPS-induced cell apoptosis were attenuated. With RT-PCR and Western blot analysis, up-regulated expressions Bcl-2 were observed.Conclusion Vitexin could protect against LPS-induced INS-1 cells injury and apoptosis,and it could be related with the regulation on the expression of Caspase-3 and Bcl-2.
