摘要
目的优选槟榔免煎颗粒提取工艺。方法以出膏率和槟榔碱含量为指标,采用正交试验和单因素试验考察槟榔药材浸泡时间、固液比、提取时间、提取次数对槟榔提取工艺的影响,通过高效液相色谱法测定槟榔碱的含量,SCX-强阳离子交换树脂柱(4.6 mm×250 mm,5μm);乙腈-磷酸溶液(2→1000)(70:30),检测波长为215 nm,流速1 mL/min,进样量10μL。结果槟榔免煎颗粒的最优提取工艺为药材浸泡2 h,加6倍量水,1h/次,煎煮2次;平均出膏率为22.69%(RSD为1.67%),槟榔碱平均含量为4.63 mg/g(RSD为1.82%)。结论优选的提取工艺稳定、可行,为槟榔免煎颗粒的开发提供了实验基础。
Objective To optimize extraction process for boil-free granule of areca catechu.Methods With the extraction amount and content of arecoline as indexes,single factor analysis method was used to develop soak time of areca catechu,and effects of ratio of solid-liquid,extraction times,extraction quantity on extraction technology were investigated by orthogonal test.The content of arecoline was determined by HPLC,SCX- strong cation chromatographic column(4.6 mm×250 mm,5 μm),acetonitrile-phosphoric acid(2→1000)(70:30),λ=215 nm,v=1 mL/min,sample quantity was 10 μL.Results The best extraction process for boil-free granule of areca catechu was as following,soak time was 2 h,ratio of solid-liquid was 6:1,extraction time was 1 h,extraction quantity was 1.Average extraction rate was 22.69%(RSD=1.67%),average content of arecoline was 4.63 mg/g(RSD=1.82%).Conclusion This optimized extraction process is stable and feasible,which can provide experimental basis for development of areca catechu boilfree granule.
