桑叶中黄酮类和酚酸类成分的提取工艺优化 被引量:4

摘要 目的:对桑叶中黄酮类和酚酸类的提取工艺进行优化,以期为该类化学成分的资源化利用提供参考。方法:采用乙醇溶剂提取桑叶中黄酮类和酚酸类成分;采用正交试验对提取工艺参数进行优化;采用HPLC-DAD方法对其中3个黄酮类化学成分和3个酚酸类成分进行分析评价。结果:各因素对黄酮类成分提取率影响的程度排序为乙醇浓度>料液比>提取时间>提取次数;各因素对酚酸类成分提取率影响的程度排序为提取时间>提取次数>料液比>乙醇浓度。通过HPLC-DAD分析其中6个资源性化学成分的评价结果及方差,黄酮类、酚酸类成分的最佳提取工艺参数:乙醇浓度为85%,料液比为1∶40,提取时间为45 min,提取次数为3次。结论:该提取工艺流程简单,提取率高,可为桑叶资源的开发利用提供参考。 Objective:The study is aimed to optimize the extract parameters of flavonoids and phenolic acids from mulberry leaves in order to provide the basis for the resource utilization.Method:The ethanol solvent was selected to extract the flavonoids and phenolic acids from mulberry leaves.And the extraction parameters were optimized by orthogonal experiment and HPLC-DAD method was adopted to analyze the flavonoids and phenolic acids.Results:The results showed that the influence degree of various factors for flavonoids is:ethanol concentration〉ratio of material to liquid〉extraction time〉extraction frequency.For phenolic constituents,the extraction frequency〉extraction time〉ratio of material to liquid〉ethanol concentration.Based on the six bioactive compositions and variance analysis data,the optimal parameters of the extraction for flavonoids and phenolic acids were:ethanol concentration 85%,ratio of material to liquid was 1∶40,extraction time is 45 minutes,3 times of extraction,respectively.Conclusion:The extraction process of flavonoids and phenolic acids from mulberry leaves has the advantages of simplicity,high extraction rate,and it provides the basis and reference for the development and utilization of mulberry resources.
