摘要
为了建立复方蒲公英颗粒的可控质量标准,建立了同时测定复方蒲公英颗粒中秦皮乙素和咖啡酸含量的高效液相色谱法,并测定样品中两种成分的含量。采用Wonda Cract ODS-2(250 mm×4.6 mm,5μm)色谱柱,以甲醇-0.6%冰醋酸水溶液(20∶80)为流动相,体积流量为1.0 m L/min,检测波长为326 nm,柱温为25℃。结果表明,秦皮乙素和咖啡酸分别在0.02~0.32 mg/m L和0.004~0.064 mg/m L浓度范围内与峰面积呈现良好的线性关系(R^2=0.9999),平均加样回收率分别为100.08%和99.69%,RSD分别为0.98%和0.84%。本方法准确、灵敏、可靠,重现性好,可用于复方蒲公英颗粒产品的质量控制。
In order to improve the quality standard of compound dandelion granule,an HPLC method was established for simultaneously determination of aesculetin and caffeic acid in compound dandelion granule. The isolation was performed on a Wonda Cract ODS- 2 column( 4. 6 mm × 250 mm,5 μm) with mobile phase consisting of methanol – 0. 6% glacial acetic acid( 20 ∶ 80). The flow rate was 1. 0 m L / min,the detection wavelength was 326 nm and the column temperature was 25 ℃. The results showed that good linear relationship was obtained in the range of 0. 02 ~ 0. 32 mg / m L and 0. 004 ~ 0. 064 mg / m L respectively( R2= 0. 9999). The average recoveries of aesculetin and caffeic acid were 100. 08% and 99. 69% with RSD of 0. 98% and 0. 84%.The method was accurate,sensitive and reliable with a good reproducibility and can be used as an quantitative analysis of aesculetin and caffeic acid in compound dandelion granule.
