胃癌相关长链非编码RNA的生物信息学筛选 被引量:5

摘要 目的用生物信息学方法筛选胃癌中差异表达的长链非编码RNA。方法于NCBI公共数据平台GEO下载胃癌基因芯片数据GSE33335,用SAM软件对其进行差异表达分析,对差异表达基因进行重注释并筛选其中的长链非编码RNA,用Gene Cluster和Tree View软件对筛选出的长链非编码RNA的表达情况予以验证。结果针对该芯片数据,筛选出胃癌中差异表达的非编码RNA 15个,其中,表达上调的12个,表达下调的3个,这15个非编码RNA表达值倍数均>2或<0.5(q均<0.05)。结论用生物信息学方法筛选胃癌相关的长链非编码RNA是一种非常高效的手段,能为胃癌相关肿瘤标志物的探索提供理论依据。 Objective To investigate the differential expression of long non-coding RNAs( lncRNAs) in gastric cancer by bioinformatics approach. Methods The microarray data of gastric cancer GSE33335 was downloaded from Gene Expression Omnibus( GEO),the public data platformin of NCBI. The differentially expression was analyzed with SAM software. The genes with differentially expression were re-annotated and the long non-coding RNA was screened. The data of the long non-coding RNA exptrssion were validated by Gene Cluster and Tree View software. Results Based on the data from the microchip 15 differentially-expressed lncRNA in gastric carcinoma tissue were screened,among which 12 were up-regulated and 3 were down-regulated. All the lncRNA showed notably differential expression and the fold-change of gene expression was more than 2 or less than 0. 5( all q 〈0. 05). Conclusion The bioinformatics approach may provide a novel and efficient way to screen cancer-associated lncRNA as the new biomarkers for further study on gastric carcinoma.
