Electrophoretic patterns of the AP-PCR products from stomach and its cancer tissues are of polymorphic characteristics

摘要 为了研究细胞癌变后DNA结构的改变 ,探讨这种改变和肿瘤发生、发展的关系 ,用AP -PCR扩增胃癌及其癌旁和正常组织DNA。结果表明 ,引物P1 ( 5′CGGCCCCGGT3′)对上述三种组织的扩增产物的电泳图谱表现出多态性 ;然而 ,引物P2 ( 5′TTTGC CCGGT3′)对上述三种组织的扩增产物的电泳带型一样 ,没有表现出多态性。这说明 ,细胞癌变后DNA结构在引物P1顺序处发生了突变 ; In order to study the alteration of DNA structure along with the cancerization of cells and search for the tumor-specific DNA fragments or tumor-related genes, AP-PCR was used to amplify the DNA samples of stomach cancer tissues and its paracarcinoma tissues or normal tissues. The electrophoretical patterns of the products amplified from the above tissues with primer P1(5′GGCCCCGGT3′) were of polymorphic characteristics. The stomach and its paracarcinoma tissues gave out six electrophoretic bands, while no electrophoretic band was amplified from stomach cancer tissue by prine P1. However, the products amplified from the above tissues with primer P2 (5′TTTGCCCGGT3′) gave out the same electrophoretic patterns. This suggests that mutations happened at the sites of the sequence 5′CGGCCCCGGT3′on DNA along with the cancerization of stomach cells.
