摘要
目的考察中药复方(Chinesherbal compound,CHC)配伍酪蛋白水解物(Casein hydrolysate,CH)后对复方降血压作用的影响。方法 1)采用小鼠最大耐受量考察中药复方-酪蛋白混合物(CHC-CH)的急性毒性。2)SD大鼠分为空白对照组和正常给药组,分别给予蒸馏水(10 ml/kg)和CHC-CH(0.67 g/kg,0.0.033 g/kg);原发性高血压大鼠(SHR)分为模型组,牛黄降压胶囊组(0.288g/kg),低、中、高剂量CHC与CH配伍组(低剂量CHC-CH分别为0.33和0.016 g/kg、中剂量CHC-CH分别为0.67和0.033 g/kg,高剂量CHC-CH分别为1.33和0.067 g/kg)和CHC组(0.033 g/kg),每天灌胃给药1次,连续给药4周,于第1、2、4周及停药后1周记录各组大鼠收缩压、舒张压和心率,动物处理后测定血清一氧化氮(NO)、血管紧张素Ⅱ(AngⅡ)、内皮素-1(ET-1)含量。结果 1)CH-CHC的最大给药剂量为96 g生药/kg合并CH 5.6 g/kg,并未见引起小鼠明显急性毒性反应。2)与高血压模型对照组比较,CHC-CH组和CHC组SHR大鼠的收缩压和舒张压、NO、AngⅡ、ET-1含量等均具有显著性差异(P<0.05或P<0.01),CH-CHC降血压作用显著大于CHC组(P<0.05),各药物组对大鼠心率影响均无显著差异。结论该中药复方与酪蛋白水解物配伍后未见具有明显急性毒性。中药复方具有显著的降血压作用,配伍酪蛋白水解物可以加强其降压作用,两者对正常动物血压无影响。
Objective To observe the influence of casein hydrolysate( CH) to Rhizoma Gastrodiae, Eucommia Ulmoides, Polygonium Multiflorum and Radix Angelicae Dahuricae compound( Chines herbal compound, CHC) on antihypertensive effect and acute toxicity.Methods KM rats were divided into control group and CH-CHC groups, distilled water and CH incorporated with CHC(CH-CHC) were intragastric administered at the Maximum gastric capacity, twice every day for 14 days. SD rats were divided into control group and normal control group, distilled water and CHC-CH, were intragastric administered. SHR rats were divided into positive control group and 5experiment groups, Niuhuang Jiangya capsules(0.288g/kg), CHC-CH low dose group, low, middle, high dosage of CH-CHC and CHC group, and all groups were daily intragastric administered respectively for 4 weeks. Systolic pressure( SP), diastolic pressure( DP),heart rate(HR), angiotoninⅡ(AngⅡ), nitric oxide(NO) and endothelin-1(ET-1) were determined. Results SP, DP, Ang, NO and ET-1 of rats from CHC-CH groups and CHC group were all decreased significantly comparing with those of control groups(P〈0.05 or P〈0.01). SP, DP and AngⅡlevels of serum were also significant decrease between CHC-CH and CHC groups, the normal controle group had no significant different with control group. The HR of every group and all itmes in acute toxcity test had no significant different.Conclution CHC-CH and CHC groups had significant antihypertensive effects on SHR rats, and CH can enhance the effects significantly,which had no influence on hypotension on normal rats and no acute toxicity.
