Expressions of CDC7 and MCM4 in endometrial adenocarcinoma detected by RT-PCR

摘要 目的研究细胞分裂周期蛋白7(CDC7)和微小染色体维持蛋白4(MCM4)两者mRNA在子宫内膜腺癌及正常子宫内膜之间的差异性表达,并探讨CDC7和MCM4与子宫内膜腺癌的关系。方法收集新鲜正常子宫内膜组织标本40例,新鲜子宫内膜腺癌组织标本100例,以β-actin为内参,运用RT-PCR技术分别检测CDC7和MCM4的mRNA表达情况。结果CDC7和MCM4的mRNA在子宫内膜腺癌中的表达量明显高于正常子宫内膜组织,差异有统计学意义(P<0.05);两者mRNA的表达与浸润肌壁深度、FIGO分期、分化程度及淋巴结转移有显著关系(P<0.05),但与年龄无关(P>0.05)。结论 CDC7和MCM4的表达与子宫内膜腺癌的发生、发展有一定的相关性,两者的基因表达可能与异常细胞周期有关。 Objective To research the differential expressions of cell division cycle protein 7 (CDC7) and minichromosnme main- tenance protein 4 (MCM4) mRNA in endometrial adenocarcinoma and normal endometrium, explore the relationships between CDC7, MCM4 and endometrial adenocareinoma. Methods Forty fresh and normal endometrial specimens and one hundred fresh endometrial adenocarcinoma specimens were collected, RT-PCR was used to detect the expressions of CDC7 and MCM4 mRNA taking 13-actin as internal reference. Results The expression levels of CDC7 and MCM4 mRNA in endometrial adenoearcinoma were statistically significantly higher than those in normal endometrium ( P〈0.05 ) . The expressions of CDC7 and MCM4 mRNA were significantly correlated with the depth of myometrial in- vasion, FIGO stage, differentiated degree, and lymph node metastasis ( P〈0. 05 ) , which were not correlated with age ( P〉0. 05 ) . Conclu- sion The expressions of CDC7 and MCM4 are correlated with occurrence and development of endometrial adenocarcinoma to a eertain extent, which may be related to abnormal cell cycle.
