摘要
【目的】通过transwell趋化实验,观察经慢病毒转染后过表达CXCR4的人脐带间充质干细胞(human umbilical cord mesenchymal stem cells,HUMSCs)是否具有更好的迁移作用。【方法】通过组织贴壁法,分离和培养来源于人脐带华通胶组织中的MSCs。经慢病毒转染,将CXCR4目的基因转染至第3代HUMSCs,并进行免疫荧光、western blotting检测在HUMSCs中表达CXCR4基因的情况。通过transwell小室进行HUMSCs的趋化迁移试验检测,观察转染CXCR4基因后的HUMSCs迁移能力是否增强。【结果】自脐带分离培养的HUMSCs通过慢病毒转染后,可过表达CXCR4基因。Transwell试验发现,过表达CXCR4的HUMSCs较正常HUMSCs和未转染目的基因的HUMSCs,迁移的细胞数明显增加(P<0.05)。【结论】通过慢病毒转染过表达CXCR4的HUMSCs,其趋化、迁移能力明显增强。
【Objective】To observe the effect of overexpressing CXCR4 by gene modified on migration capacity of HUMSCs by transwell test.【Methods】MSCs were isolated from Wharton's jelly of human umbilical cords by means of adherent culture. HUMSCs were infected by lentivirus constructed with CXCR4. The expression of CXCR4 gene was examined through immunofluorescence and western blot. The migration capacity of overexpressing CXCR4 HUMSCs was verified by transwell test.【Results】After infected by lentivirus constructed with CXCR4, HUMSCs showed overexpression of CXCR4 protein. And it's migratory cells amount was more than normal and control HUMSCs respectively.【Conclusion】Through gene modified, the migration capacity of overexpressing CXCR4 HUMSCs was increased significantly.
