复方苦参注射液对rBMSCs细胞增殖和迁移能力的影响 被引量:3

摘要 目的采用不同浓度复方苦参注射液在常氧或缺氧条件下作用不同时间后,观察rBMSCs细胞增殖和迁移能力的变化情况。方法 MTT法检测rBMSCs增殖的情况;ELISA法检测rBMSCs培养上清中SDF-1水平的变化;Transwell检测rBMSCs迁移能力的改变。结果在100μL/mL、200μL/mL和300μL/mL组处理24h和48 h后细胞的存活率,组间比较差异有统计学意义(P<0.05)。在200μL/mL处理24 h组,细胞上清液中SDF-1含量上升,差异有统计学意义(P<0.05)。200μL/mL组细胞迁移率与其他剂量组比较,差异有统计学意义。结论在适当浓度复方苦参注射液作用下,在低氧条件下能提高rBMSCs细胞存活率,能促进rBMSCs分泌SDF-1,利于rBMSCs细胞发生迁移。 Objective To observe the effect of compound radix sophora flavescentis injection under the condition of normoxia and hypoxia on rBMSCs of survival and migration ability. Methods MTT assay was applied to measure the survival of rBM- SCs in vitro. ELISA was used to determine the levels of SDF-1 protein in the culture supematant of rBMSCs. By using Transwell assay, the migration rate of rBMSCs was observed. Results The cell survival rate was statistically significant in 100 μL/mL, 200 μL/mL and 300 μL/mL group for 24 h and 48 h ( P 〈 0.05). In 200 txL/mL group for 24 h compared with other groups, SDF-1 in the cell culture supematant increased, with statistical significance (P 〈 0.05). Cell migration rate of 200 ~L/mL group compared with other groups, the difference was statistically significant. Conclusion Appropriate concentration compound radix sophora flavescentis injection under the condition of hypoxia can improve rBMSCs survival rate, promote rBMSCs cells to secrete SDF-1, and conduce rBMSCs cell migration.
