双环醇改善荷瘤动物生存状态的实验研究

摘要 目的双环醇单用及与抗肿瘤药物合用对小鼠生存状态的影响。方法昆明种小鼠皮下接种S180肉瘤细胞悬液后第6天开始灌胃给予双环醇(200 mg·kg-1×7),并在给药第1天和第6天,分别腹腔注射阿霉素(8 mg·kg-1×2)、环磷酰胺(70 mg·kg-1×2),于末次给药后24 h,随机取6只动物剥瘤称重,其余动物记录死亡率。C57BL/6小鼠皮下接种Lewis肺癌后第9天开始灌胃给予双环醇(150 mg·kg-1×14),观察动物状态并记录死亡情况。C57BL/6小鼠皮下接种B16-F10黑色素瘤后第5天开始灌胃给予双环醇(50 mg·kg-1×18,100 mg·kg-1×18),观察动物状态并记录动物死亡情况。实验过程中同时设抗肿瘤药物对照组和荷瘤空白对照组。结果双环醇单用在荷S180肉瘤小鼠模型和荷B16-F10黑色素瘤小鼠模型均能显著延长动物的中位生存期,提高实验周期内动物的存活率。在荷Lewis肺癌小鼠模型,双环醇未显示良好的延长中位生存期的活性。双环醇与阿霉素或环磷酰胺合用,均对后者的抑瘤活性有略增加的作用。双环醇与阿霉素合用能进一步延长动物的中位生存期,提高实验周期内动物的存活率。双环醇与环磷酰胺合用,无进一步改善小鼠生存状态的作用。结论双环醇单用或与抗肿瘤药物合用,有延长荷瘤动物中位生存期,改善动物生存状态的活性,值得进一步关注。 Objective To investigate the effect of bicyclol and combinative effect of bicyclol with anti-tumor drugs on survival condition of tumor-bearing mice. Methods Kunming mice transplanted with S180 sarcoma were administrated with 200 mg·kg^-1 bicyclol for 7 days, and simultaneously injected with 8 mg·kg^-1 adriamycin or 70 mg·kg^-1 cyclophosphamide at the first day and sixth day. After treatment, six mice of each group were sacrificed and the inhibitory rate of tumor growth was calculated. Animal death in the remaining animals was recorded. C57BL/6 mice were administrated with 150 mg·kg^-1 bicyclol for 14 days after 9 days transplanted with Lewis lung carcinoma. C57BL/6 mice were treated with bicyclol (50 mg·kg^-1,100 mg·kg^-1) for 18 days after 5 days transplanted with B16-F10 melanoma carcinoma. In these two models, the body weight and survival condition of mice were observed. Results In S180 sarcoma and B16-F10 melanoma tumor-bearing mice models, bicyclol significantly prolonged the median survival time and decreased the mortality rate of tumor-bearing mice. However, there was no marked improvement effect at mice survival rate in C57BL/6 Lewis lung tumor-bearing mice models with bicyclol treatment. The combination of bicyclol with anti- tumor drugs showed slight increase on their anti-tumor activities. Combinedapplicadon with bicyclol and adriamycin, bicyclol could also prolong the median survival time and decrease the death rate of mice induced by adriamycin. Bicyclol combined with cyclophosphamide could not effectively improve the living conditions of mice. Conclusion Bicyclol administration or combination therapy with anti-tumor drugs can prolong the median survival time of tumor-bearing mice and obviously improve the mice living conditions.
