摘要
目的:观察丹红注射液对血管性痴呆大鼠学习记忆能力的影响,并探讨其作用机制。方法:将SD大鼠随机分为假手术组、对照组和丹红组,每组又分为6h、24h、72h、1w、1m(n=6)四个亚组。采用双侧颈总动脉永久性阻断法(2-VO)建立VD大鼠模型,暗回避反应法测定学习记忆能力;应用免疫组织化学法、原位末端标记法检测大鼠海马CA1区HIF-1α表达及凋亡细胞数的变化。结果:12-VO术后72h凋亡细胞数最多,与对照组相比,丹红组各时间点凋亡细胞数目显著减少;26h时丹红组潜伏期与对照组比较无统计学意义,从24h开始丹红组各时间点潜伏期显著延长;36h时丹红组HIF-1α蛋白表达与对照组无明显统计学差异,24h丹红组表达明显升高,而从72h之后HIF-1α蛋白表达显著降低。结论:丹红注射液对血管性痴呆大鼠学习记忆障碍具有改善作用,其机制可能与调节了HIF-1α蛋白表达,减少了海马CA1区的细胞凋亡有关。
Objective:To observe the effect of Danhong Injection on learning-memory disorder of vascular dementia rats,and to explore its mechanism.Methods:Male SD rats were randomly divided into three groups:shamoperated group,control group and Danhong group.The vascular dementia model was created by apermanent bilateral common carotid arteries occlusion(2-VO)in Sprague-Daewley rats.Rats were evaluated on learning-memory ability by passive avoidance test.At corresponding time points at 6h,24 h,72h,1wand 1m,the dynamic changes on the cell counts of HIF-1aand apoptosis in hippocampus CA1 were determined by immunohistochemical method and TUNEL respectively.Results:1The apoptotic neurons were injured most severely at 72 hin control group.While in the Danhong group,the number of neuronal apoptosis significantly decreased at respective time.2Compared with the rats in control group,the performance on passive avoidance test of rats in Danhong group was improved significantly after 24 h.There were no differences at 6h.3The level of positive expression of HIF-1αin the hippocampal CA1 significantly increased at 24 h,but significantly decreased after 72 h.Conclusion:Danhong Injection could improve the learning-memory ability of vascular dementia rats.The protective mechanism might be related to the effect of preventing apoptosis in rat hippocampus CA1 and regulating expression of HIF-1a.
