摘要
目的机体损伤和病变造成的组织缺损通常可以组织工程的方式来进行修复。方法本研究结合细胞片层技术和静电纺丝技术,利用脂肪干细胞制备多细胞片层,并对各种电纺生物膜结构形貌、亲水性能、力学性能、以及与脂肪干细胞相容性等进行较为深入的比较研究。结果表明制备的仿细胞外基质(ECM)结构的多孔网状电纺生物膜在形态上模拟ECM,而且具有孔隙率高,贯通性好,比表面积高,利于细胞的生长和粘附的特点。结论本研究利用电纺生物膜作为支撑膜,增加了细胞的增殖空间和营养物质的流通,又保证了细胞片层在转移过程中的完整性,从而给临床多细胞片层移植治疗提供了一个简便易行的操作方法。
Objective To explore whether tissue defects caused by body damage and lesions can be repaired through tissue engineering. Methods By integrating techniques of cell sheet and electrospinning, multiple cell sheets were made with adipose-derived stem cell(ADSC). The prepared biofilms were studied and compared for structural morphology,hydrophilic properties,mechanical properties and compatibility with ADSCs. Results The prepared electrospun biofilms showed a porous network structure which morphologically mimicked extracellular matrix (ECM). The biofilms were featured by high porosity, penetration resistance and high specific surface area which will help cell growth and adhesion. Conclusion he cell sheet prepared in this study is supported by electrospun biofilm with sufficient space for proliferation space and nutrients transportation. These features ensure the integrity of cell sheet during the transfer process,and thus offering a simple and easy method for clinical transplantation of multiple cell sheets.
