四君子汤对脾气虚证大鼠脑肠CaM/CaMKⅡ干预效应研究 被引量:16

Study on interference effect of Sijunzi decoction on brain-gut CaM / CaMKⅡ of spleen Qi deficiency syndrome rats

摘要 目的:观察脾气虚证大鼠脑肠中CaM信号通路关键基因的时相性动态表达及四君子汤干预作用。方法:Wistar雄性大鼠随机分为正常对照组、模型组(14,21,28 d)、四君子汤组(14,21,28 d),除对照组外,其余各组均以苦寒破气法(大黄枳实厚朴制剂液7.5 g·kg-1·d-1)、游泳力竭法双因素法复制脾气虚证模型,各四君子汤组造模7 d后,继续造模的同时,以四君子汤20 g·kg-1·d-1灌胃进行干预治疗,采用免疫组织化学方法、蛋白免疫印迹技术分别检测大鼠海马、小肠在不同时间阶段CaM信号转导通路关键基因CaM/CaMKⅡ表达水平,同时检测以益气健脾法为指导的四君子汤对脾气虚证大鼠的干预效应,并分析其作用机制。结果:脾气虚证大鼠相对于正常大鼠,小肠组织中CaM/CaMKⅡ表达升高,海马组织中CaM/CaMKⅡ表达降低(P<0.05,P<0.01)。四君子汤治疗后大鼠相对于脾气虚证大鼠,小肠组织中CaM/CaMKⅡ表达明显降低,海马组织中CaM/CaMKⅡ表达明显升高(P<0.05,P<0.01)。结论:脾气虚证证候的形成有可能与CaM/CaMKⅡ在小肠组织中的高表达、海马组织中的低表达有关,四君子汤对脾气虚证的治疗作用有可能是通过降低小肠组织并升高海马组织中CaM/CaMKⅡ的表达来实现的。 Objective: To observe the dynamic time-phase expressions of key genes of brain-gut CaM signal pathway of spleen Qi deficiency rats and the intervention effect of Sijunzi decoction. Method: Male Wistar rats were randomly divided into the normal control group,model 14 d,21 d,28 d groups,and Sijunzi decoction 14 d,21 d,28 d groups. Except for the normal control group,the remaining groups were included into the spleen Qi deficiency model with the bitter cold breaking Qi method( ig 7. 5 g·kg^-1·d^-1of Rheum officinale,Fructus aurantii immaturus,Magnolia officinalis preparation) and the exhaustive swimming method. On the 7thday after the modeling,the Sijunzi decoction groups were orally administered with Sijunzi decoction 20 g·kg^-1·d^-1. The expressions of key genes CaM / CaMKⅡ of CaM signaling pathway in hippocampus and intestine at different time points by immunohistochemical method and Western blot. At the same time,the intervention effect of Sijunzi decoction on spleen Qi deficiency rats and its mechanism were analyzed. Result: Spleen Qi deficiency rats showed higher intestinal CaM / CaMKⅡ expression and lower hippocampus CaM / CaMKⅡexpression than normal rats( P〈0. 05,P〈0. 01). After the treatment of Sijunzi decoction,spleen Qi deficiency rats showed reduction in intestinal CaM / CaMKⅡ expression and increase in hippocampus CaM / CaMKⅡ expression( P〈0. 05,P〈0. 01). Conclusion:The formation of spleen Qi deficiency syndrome may be related to the high expression of CaM / CaMKⅡ in small intestine tissues and its low expression in hippocampus tissues. Sijunzi decoction may achieve the therapeutic effect in spleen Qi deficiency syndrome by reducing the CaM / CaMKⅡ expression in intestinal tissues and increasing it in hippocampus tissues.
