摘要
目的:观察海蛇药酒联合甲氨蝶呤对胶原诱导关节炎大鼠血清CD4/CD8及Ig A、Ig M、Ig G的影响,研究其对胶原诱导关节炎的治疗作用。方法:将Wistar雄性随机分4组:正常对照组、模型组、MTX给药组、海蛇药酒+MTX给药组,每组12只,建立II型胶原诱导大鼠类风湿性关节炎模型,第二次加强免疫一周后,对照组给予生理盐水灌胃,各给药组分别给予对应药品,连续给药两个月,各组大鼠经腹主动脉取血,血液离心后留上清液,应用流式细胞仪检测大鼠血清CD4、CD8,采用ELISA法检测大鼠中血清Ig A、Ig M、Ig G含量。结果:(1)各组大鼠血清CD4、CD8、CD4/CD8数值:与模型组比较,对照组和各治疗组CD4数值有所降低,但无显著性差异(P>0.05);对照组和各治疗组CD8数值有所升高,且有显著性差异(P<0.05);对照组和各治疗组CD4/CD8数值均显著降低(P<0.05)。海蛇药酒+MTX组与MTX组组间比较,CD4数值有所降低,但无显著性差异(P>0.05);CD8数值亦有降低,且有显著性差异(P<0.05);CD4/CD8比值降低,有显著性差异(P<0.05)。(2)各组大鼠血清Ig M、Ig A、Ig G水平:与模型组比较,对照组与各治疗组大鼠Ig M、Ig A、Ig G水平均显著降低(P<0.05)。联合给药组大鼠Ig M、Ig A、Ig G水平显著优于MTX组,且与MTX均有显著性差异(P<0.05)。结论:使用海蛇药酒联合MTX效果优于单纯使用MTX,其机理可能通过纠正CD4/CD8的失衡而调节RA免疫功能紊乱、恢复细胞免疫动态平衡,维持CD4/CD8的动态平衡,并通过抑制免疫球蛋白和RF的分泌,抑制血管翳的形成、B细胞合成急性异蛋白,由此抑制炎症反应,最终起到治疗关节炎的目的,但仍需要进一步的研究与验证。
Objective: To observe sea snake wine combined with methotrexate( MTX) serum on collagen- induced arthritis rats' CD4/ CD8 and Ig A,Ig M,Ig G and study its therapeutic effect on collagen- induced arthritis. Methods: The Wistar male rats were randomly divided into four groups: normal control group,model group,MTX treatment group,sea snake wine + MTX group,12 for each. We established type II collagen- induced arthritis model in rats. One week after the second booster immunization,the control group received normal saline and treatment groups were given the corresponding drugs,continuously for two months. Each group's abdominal aorta blood was taken out,and then leaving the supernatant after centrifugation. The flow cytometry was used to detect serum levels of CD4 and CD8and ELISA to detect the serum Ig A,Ig M and Ig G contents. Results:( 1) Each group' s serum CD4,CD8,CD4/ CD8value: Compared with the model group,the control group and the treatment groups decreased CD4 values,with no significant difference( P〉 0. 05); control group and each treatment group' s CD8 value increased,and there was a significant difference( P 〈0. 05); control group and each treatment group's CD4/ CD8 values were significantly lower( P 〈0. 05). Sea snake wine + MTX group and the MTX group's CD4 value decreased,but there was no significant difference( P〉0. 05); CD8 value also decreased,and there was a significant difference( P 〈0. 05); CD4/ CD8 ratio was lower and there was a significant difference( P 〈0. 05).( 2) Each rat's serum Ig M,Ig A and Ig G levels: Compared with the model group,the control group and each treatment group's Ig M,Ig A and Ig G levels were significantly lower( P 〈0. 05). The combination administration group' s Ig M,Ig A and Ig G levels were significantly superior to that of the MTX group,with significant differences( P〈 0. 05).
