国内不同产地姜黄药材的指纹图谱分析与含量测定 被引量:10

摘要 本文搜集全国各产地的姜黄药材共17批次样品,以甲醇超声提取60 min,以YMC-Pack-ODS-A C18(250 mm×4.6 mm i.d.,5μm)为分离色谱柱,乙腈-0.1%甲酸溶液为流动相,流速1.0 m L/min,柱温30℃,分别在检测波长270 nm下进行指纹图谱分析,380 nm下进行含量测定分析。结果表明,17批不同产地的姜黄药材在化学成分组成上基本一致,共检测到P1~P6 6个共有峰。不同产地药材中主要成份姜黄素类化合物的含量具有显著差异。检测到姜黄素的含量在4.18~22.28 mg/g;脱甲氧基姜黄素含量在1.08~9.26 mg/g;双脱甲氧基姜黄素含量在0.40~9.50 mg/g。本试验表明,1本文建立的基于HPLC的定性指纹图谱研究及定量含量测定研究方法可用于对姜黄药材进行质量控制;2不同产地姜黄药材的化学组成一致,但是姜黄素类化合物的含量差异较大,在道地产区四川种植的姜黄药材中,总姜黄素的含量显著高于其他产区。 In this paper,17 batches of samples were respectively collected from different cultivation areas,which were extracted by methanol in ultrasonic bath for 60 min,detected for fingerprint at 270 nm and quantitatively analyzed at 380 nm with HPLC method of a YMC-Pack-ODS-A C18 column,gradient elution of CH3 CN and 0. 1 % formic acid at a flow rate of 1. 0 m L / min,a column temperature of 30 ℃ and UV. The result showed that the HPLC fingerprint analysis exhibited that the chemical composition of 17 batches of Curcuma longa samples from different areas were identical,and P1-P6 were detected as common peaks. The contents of three curcuminoids from different Curcuma longa samples were significantly different; The content of curcumin ranged from 4. 18- 22. 28 mg / g; The content of demethoxycurcumin ranged from 1. 08- 9. 26 mg / g; And the content of bisdemethoxycurcumin ranged from 0. 40- 9. 50 mg / g. It was concluded that( i) The validated HPLC fingerprint and quantitative method could be used for quality control of Curcuma longa herbal medicines.( ii) The chemical composition of Curcuma longa from different areas were highly similar,but the content of three curcuminoids was different significantly; among all the Curcuma longa samples analyzed,the highest contents of total curcuminoids were detected from Sichuan province.
