多肽免疫抑制牙龈卟啉单胞菌引起的小鼠感染及牙槽骨吸收作用的研究 被引量:5

Inhibition of amino acid sequence of hemin-binding sites of Porphyromonasgingivalis hemagglutinin-2 against infection and alveolar bone absorption in mice

摘要 目的探讨牙龈卟啉单胞菌(Porphyromonasgingivalis,P.g)血凝素2(hemagglutinin-2,HA-2)的氯化血红素结合位点多肽作为抗原免疫小鼠,对降低P.g感染后毒性和抑制牙周炎牙槽骨吸收的作用。方法将多肽与牛血清白蛋白(Bovine serum albumin,BSA)偶联作为抗原免疫BALB/c小鼠。P.g国际标准菌株ATCC33277厌氧培养,皮下注射菌液,监测小鼠脓肿的大小及转归,评价多肽对P.g皮下感染的保护作用;口腔接种细菌感染小鼠,检测小鼠牙周炎牙槽骨吸收量,评价其对于P.g引起的牙周炎牙槽骨吸收的保护作用。结果多肽DGFPGDHYAVMISK作为抗原免疫小鼠可减小小鼠皮下接种P.g处形成脓肿的大小,并加快皮下脓肿的愈合;可抑制口腔感染P.g后小鼠牙周炎模型牙槽骨的吸收。结论多肽DGFPGDHYAVMISK免疫小鼠对P.g感染小鼠具有一定的保护作用;可减少小鼠牙周炎模型牙槽骨的吸收。为开发新的牙周病防治方法奠定基础。 Objective To discuss the effect of amino acid sequence of hemin-binding sites of Porphyromonasgingivalis(Pg)hemagglutinin-2(Pg HA-2)in reducing the Pg toxicity and inhibiting the periodontitis alveolar bone absorption in antigen peptides-immunized mice.Methods Mice were immunized with the antigen containing peptides coupled with bovine serum albumin(BSA).The standard Pg strain ATCC 33277 was cultured.For subcutaneous infection,the mice were challenged with Pg by subcutaneous injection in abdomen,then the lesion sizes were monitored daily for over 20 days to evaluate the protective effect of the peptides against Pg subcutaneous infection.For oral infection,the mice were given the suspension of Pg,and the amount of the alveolar bone absorption was measured to evaluate the protective role of the peptides against periodontitis alveolar bone absorption caused by Pg.Results The amino acid sequence DGFPGDHYAVMISK caused the sizes of abscesses in thePg subcutaneous infection groupdecreased,and the healing of subcutaneous abscesses accelerated;the absorption of alveolar bone of periodontitis mice was inhibited.Conclusion The peptide DGFPGDHYAVMISK has certain protective effect on Pg infected mice,reducing the amount of alveolar bone absorption.The study laid a good foundation for improving the prevention and treatment of periodontal disease.
